Genome-wide pathway analysis of a genome-wide association study on psoriasis and Behcet's disease

The aim of this study was to identify candidate causal single nucleotide polymorphisms (SNPs) and candidate causal mechanisms of psoriasis and Behcets's disease (BD) and to generate an SNP -> gene -> pathway hypothesis. A psoriasis genome-wide association study (GWAS) dataset that included 436,192 SNPs in 1,409 psoriasis cases and 1,436 controls of European descent and a BD GWAS dataset that contained 310,324 SNPs in 1,215 BD cases and 1,278 controls were used in this study. Identify candidate causal SNPs and pathways (ICSNPathway) analysis was applied to the GWAS datasets. ICSNPathway analysis identified 15 candidate causal SNPs and 28 candidate causal pathways. The top five candidate causal SNPs were rs1063478 (P = 1.45E-10), rs8084 (P = 2.20E-08), rs7192 (P = 5.18E-08), rs20541 (P = 5.30E-06), and rs1130838 (P = 5.65E-06), which with the exception of rs20541 [interleukin (IL)-13] are at human leukocyte antigen (HLA) loci. These candidate causal SNPs and pathways provided ten hypothetical biological mechanisms. The most strongly associated pathway concerned HLA. When HLA loci were excluded, ICSNPathway analysis provided one hypothetical biological mechanism. rs20541 (non_synonymous_coding) -> IL-13 -> dendritic cell involvement in the regulation of Th1 and Th2 development, and the GATA3 pathway. ICSNPathway analysis identified four candidate causal SNPs, eleven candidate causal pathways, and three hypothetical biological mechanisms. One of them was as follows: rs2072895 (non_synonymous_coding & splice-site) and rs2735059 (non_synonymous_coding) -> HLA-F -> type I diabetes mellitus, antigen processing and presentation, and autoimmune thyroid disease. The application of ICSNPathway analysis to GWAS dataset of psoriasis and BD resulted in the identification of candidate causal SNPs and candidate pathways that might contribute to psoriasis susceptibility.

Comparative efficacy and safety of infliximab and its biosimilars in patients with rheumatoid arthritis presenting an insufficient response to methotrexate A network meta-analysis

Summary: This study aimed to compare the relative efficacy and safety of infliximab and its biosimilars in patients with active rheumatoid arthritis (RA) who showed an inadequate response to methotrexate (MTX). A Bayesian network meta-analysis was conducted, combining direct and indirect evidence from randomized controlled trials (RCTs) to compare the efficacy and safety of infliximab biosimilars and the originator product in this patient population. The results showed that there were no significant differences in ACR20 response rates and serious adverse events (SAEs) between infliximab biosimilars and the originator.

A network meta-analysis of randomized controlled trials comparing the effectiveness and safety of voclosporin or tacrolimus plus mycophenolate mofetil as induction treatment for lupus nephritis

Summary: This study compared the effectiveness and safety of voclosporin + MMF, tacrolimus + MMF, and monotherapy for induction treatment in lupus nephritis patients. The results showed that tacrolimus + MMF and voclosporin + MMF were more effective than monotherapy, with tacrolimus + MMF being the most effective. However, tacrolimus + MMF had a higher risk of serious adverse events compared to monotherapy.

Placebo and nocebo responses in randomized controlled trials of non-tumor necrosis factor biologics and Janus kinase inhibitors in patients with active rheumatoid arthritis showing insufficient response to tumor necrosis factor inhibitors: A meta-analysis

Summary: This study evaluated the frequency and magnitude of placebo and nocebo responses in patients with rheumatoid arthritis (RA) who had an insufficient response to tumor necrosis factor (TNF) inhibitors and were treated with non-TNF biologics and Janus kinase (JAK) inhibitors. The results showed that JAK inhibitors had higher placebo effects and less nocebo effects compared to non-TNF inhibitors in this group of patients. The study also found a positive correlation between placebo response and treatment response, and a negative correlation between placebo response and adverse effects.

The lessebo effect in randomized controlled trials of rituximab in patients with rheumatoid arthritis: a meta-analysis

Summary: This study examined the impact of negative expectations associated with receiving a placebo (the lessebo effect) on efficacy outcomes in randomized clinical trials (RCTs) of rituximab in patients with rheumatoid arthritis (RA). The results demonstrated that the use of a placebo was associated with a clinically significant reduction in the magnitude of change of the ACR20, 50, 70 response rates in rituximab RCTs for RA. This has potential implications for the development of new treatments and appraisal of current treatment options for RA.

Comparative Efficacy and Safety of Biological Agents in the Treatment of Lupus Nephritis: A Network Meta-Analysis

Summary: This study assessed the relative efficacy and safety of various biological agents used in induction therapy for lupus nephritis. Associations between interleukin 17A and 17F polymorphisms and asthma susceptibility: A meta-analysis

Relative effectiveness and safety of interleukin-6 and Janus kinase inhibitors versus adalimumab in patients with rheumatoid arthritis: a network meta-analysis

Summary: This study compared the efficacy and tolerability of IL-6 and JAK inhibitor therapies with adalimumab in patients with RA and inadequate responses to MTX. The results showed that IL-6 and JAK inhibitors were more effective in achieving ACR20 response rate compared to adalimumab. However, adalimumab was associated with lower risk of serious adverse events.

Efficacy and safety of tacrolimus versus mycophenolate mofetil as induction treatment and low-dose tacrolimus as treatment for lupus nephritis: a meta-analysis

Summary: This study compared the efficacy and safety of tacrolimus and mycophenolate mofetil (MMF) as induction therapy for lupus nephritis (LN). The results showed that there was no difference between tacrolimus and MMF in terms of complete or partial remission rates. However, tacrolimus had a lower incidence of herpes zoster infection and a higher risk of serum creatinine elevation compared to MMF. Overall, 3 mg/d tacrolimus was proven to be an effective and safe treatment for LN.

Systemic lupus erythematosus and toll-like receptor 9 polymorphisms: A meta-analysis of genetic association studies

Summary: This study aimed to investigate the association between TLR9 polymorphisms and susceptibility to SLE. The results showed that the rs187084 polymorphism was associated with SLE risk in the Asian population, while the rs352139, rs352140, and rs5743836 polymorphisms were associated with SLE risk in the Asian, European, and Latin American populations, respectively.

Summary: This study investigated the association between the functional Fc gamma receptor 3A (FCGR3A) V158F and FCGR2A R131H polymorphisms and responsiveness to rituximab therapy in patients with autoimmune diseases. The meta-analysis revealed a significant association between the FCGR3A V allele and responsiveness to rituximab, while no association was found for FCGR2A R131H polymorphism.

Circulating leptin and its correlation with rheumatoid arthritis activity: a meta-analysis

Summary: This study investigated the association between leptin levels in the blood of individuals with rheumatoid arthritis (RA) and the severity of the disease. The results showed that RA patients had elevated leptin levels, which were correlated with the activity of RA. These findings suggest that leptin plays a significant role in the pathophysiology of RA.

Characterization of virus-mediated autoimmunity and the consequences for pathological process in patients with systemic lupus erythematosus

Summary: This study aimed to identify differentially expressed genes (DEGs) of systemic lupus erythematosus (SLE) using gene expression-based computational methodologies. A total of 39 upregulated genes and 57 downregulated genes were identified in SLE patients compared to healthy controls. These DEGs were implicated in immune response pathways and were also associated with various virus infections. Additionally, certain genes were found to be closely associated with the quantities of immune cells.
