Dipeptidyl peptidase-IV activity assay and inhibitor screening using a gold nanoparticle-modified gold electrode with an immobilized enzyme substrate

We report on an electrochemical biosensor for the determination of the activity of dipeptidyl peptidase-IV (DPP-IV), and on a method for screening the effect of its inhibitors. An enzyme substrate (Fc-peptide) was immobilized on the surface of a gold electrode, and double signal amplification was accomplished via an additional layer consisting of phenyl rings and gold nanoparticles. The activity of DPP-IV was determined at levels as low as 39 nU center dot mL(-1) and over a linear detection range as wide as from 0.5 mu U center dot mL(-1) to 2.5 mU center dot mL(-1). The inhibitory effects of diprotin A and the His-Leu dipeptide on the activity of DPP-IV also were tested and gave IC50 values of 93.5 and 95.5 mu M, respectively. The assay is rapid, precise and selective. It may be extended to other peptidases and, possibly, proteases and their inhibitors.

MXene Coupled with CRISPR-Cas12a for Analysis of Endotoxin and Bacteria

Summary: In this study, it was found that MXene can efficiently adsorb single-stranded DNA and be coupled with CRISPR-Cas12a for sensitive detection of LPS and bacteria. Through carefully designed aptamers, the trans-cleavage activity of CRISPR-Cas12a is activated, allowing for selective and sensitive quantification of targets in different samples. This method provides a new direction for the exploration of universal analytical methods based on MXene coupled with CRISPR-Cas12a.

Identification of dual therapeutic targets assisted by in situ automatous DNA assembly for combined therapy in breast cancer

Summary: The study introduced a novel method for simultaneous identification of dual therapeutic targets in breast cancer using in situ automatous DNA assembly reaction and electrochemical techniques. The method demonstrated high sensitivity and specificity in identifying co-existed therapeutic targets, even in serum samples, and could potentially provide valuable information for enhancing the efficiency of combined therapy in clinic. The use of quantum dots as electrochemical labeling allowed for the creation of increased electrochemical signaling to reveal the co-expression of the two targets.

Chemoselective ligation assisted DNA walker for analysis of double targets

Summary: A chemoselective ligation assisted DNA walker with input and output of double signals has been developed using oxime chemistry and alkyne-azide cycloaddition. This biosensor, designed for lipopolysaccharide (LPS) and 5-hydroxymethyl-2-furaldehyde (HMF) targets, can sensitively differentiate and analyze both targets with one-to-one mapping functionality. This work offers a new perspective on analyzing double targets based on chemoselective ligation assisted DNA walker.

Boronic Ester-Mediated Dual Recognition Coupled with a CRISPR/ Cas12a System for Lipopolysaccharide Analysis

Summary: In this work, boronic ester-mediated dual recognition coupled with a CRISPR/Cas12a system has been proposed for highly specific and sensitive detection of lipopolysaccharide (LPS). The method utilizes boronic acid and LPS aptamer (LPSA) for simultaneous recognition, and CRISPR/Cas12a for signal amplification. The method has a linear detection range from 0.05 to 5000 ng/mL with a lowest detection limit of 44.86 pg/mL, and can be developed as a universal platform for the analytical detection of other carbohydrates.

Histostar-Functionalized Covalent Organic Framework for Electrochemical Detection of Exosomes

Summary: Covalent organic frameworks (COFs) have attracted much attention for their unique physical-chemical characteristics and potential applications in biosensing. In this study, spherical COFs with regular shape and good dispersion were synthesized, and a novel nanoprobe was developed by modifying Histostar on the COFs surface. The nanoprobe was then applied to fabricate an electrochemical biosensor for exosomes detection, achieving a low detection limit and wide linear detection range. This work offers a promising platform for ultrasensitive detection of exosomes.

Ordered Labeling-Facilitated Electrochemical Assay of Alpha-Fetoprotein-L3 Ratio for Diagnosing Hepatocellular Carcinoma

Summary: In this work, an electrochemical aptasensor was designed for the high-accuracy assay of AFP-L3 ratio. The aptasensor demonstrated the ability to accurately measure the AFP-L3 ratio in a linear range of 0.0008-40 ng mL-1, with satisfactory specificity and good recoveries in complex serum samples. This aptasensor may serve as a valuable tool for the measurement of AFP-L3 ratio and have great potential for early warning of hepatocellular carcinoma in clinical application.
