Summary: This study investigated the expression of ABCB1 and intratumoral pharmacokinetics of paclitaxel in esophageal cancer patients, finding that ABCB1 expression was significantly higher in tumor tissue compared to normal esophageal tissue. Differences in ABCB1 expression and tumor histological types may partly explain why tumors respond differently to systemic treatment.

Summary: The derivative NY-2 of ZLDI-8 is a potent antitumor agent with higher activity and safety compared to the parent compound. NY-2 acts on ADAM17 and affects the Notch1 and integrin beta 1 signaling pathways, leading to inhibition of non-small cell lung cancer. In vitro and in vivo experiments demonstrate that NY-2 significantly inhibits tumor formation and metastasis without causing major toxicity.

Summary: Research has shown the significant impact of molecular charge on antibody PK, with engineered complementarity-determining regions (CDRs) of trastuzumab leading to variants with increased positive charge accumulation in tissues. Whole-body PK experiments revealed a bell-shaped relationship between systemic exposure and Fv charge, and positive charge variants exhibited superior accumulation in well-perfused organs.

Summary: Sea cucumber-derived sulfated sterols showed more significant bioactivities due to the unique structure of the sulfate group, but their absorption, pharmacokinetics, and tissue distribution remain unknown. This study investigated the absorption characteristics, pharmacokinetics, and tissue distribution of sea cucumber sterols, finding that they are absorbed quickly and mainly accumulate in the liver. These results provide insights for the development of functional foods and nutraceuticals containing sea cucumber sterols.

Summary: This study investigates the potential bias in the estimation of monoclonal antibody (mAb) concentrations in the epithelial lining fluid (ELF) using the bronchoalveolar lavage (BAL) method. It demonstrates that the unbalanced extraction of mAb and urea could lead to underestimated ELF concentrations, but the choice of lavage volume does not significantly affect the partitioning of mAb into ELF.

Summary: This study used fluorescently labeled PM to investigate its distribution and pharmacokinetics in vivo. The results showed that FITC-PM had high stability and tended to be distributed in the kidney, liver, and at lower concentrations in the heart, spleen, and lungs after tail vein injection. Pharmacokinetic analysis revealed appropriate elimination rate constant, half-life time, and clearance rate for FITC-PM. The studies of pharmacokinetics and tissue distribution provided basic information that can be used to further clarify the pharmacodynamic mechanisms of PM.

Summary: Licorice (Glycyrrhiza uralensis Fisch) is widely used in China as a food and herbal medication. The present study investigated the pharmacokinetic profile of Glycycoumarin (GCM), a major coumarin in licorice, in rats. The results showed that GCM was rapidly absorbed and transformed into its conjugated metabolites, with low bioavailability and widespread distribution in various tissues except the brain.

Summary: This study explored the pharmacokinetics and tissue distribution of Enrofloxacin and its metabolite Ciprofloxian in crayfish. Results showed higher absorption and longer elimination half-life with intramuscular administration compared to oral administration. Concentrations of ENR and CIP were higher in hepatopancreas and tissues after intramuscular administration. The study suggested a specific regimen for ENR use in crayfish based on susceptibility to strains.

Summary: Advances in antibody engineering have allowed the construction of novel molecular formats, providing new opportunities for cancer immunotherapy drug discovery, but also revealing limitations in knowledge of structure-activity relationships. By constructing a representative library and using different shapes and hinge flexibilities of IgG-like formats, it was found that one-armed antibodies have higher renal uptake compared to bivalent antibodies.
