Summary: Acute myeloid leukemia (AML) is characterized by the expansion of clonally derived hematopoietic precursors undergoing a maturation block. The study shows that YY1, a member of the noncanonical PcG complexes, is significantly overexpressed in AML and knockdown of YY1 relieves the differentiation block in AML.

Summary: This study evaluated the absolute infectious risk in 200 AML patients treated with CPX-351 and found it to have a good safety profile, with an incidence of infectious complications comparable to that of pivotal studies. The only factor significantly associated with mortality was a lack of response to CPX-351 treatment.

Bone Marrow Microenvironment Involvement in t-MN: Focus on Mesenchymal Stem Cells

Summary: Therapy-related myeloid neoplasms (t-MN) are a late complication of cytotoxic therapy (CT) used in the treatment of both malignant and non-malignant diseases. Among the factors contributing to t-MN development, the bone marrow microenvironment and bone marrow mesenchymal stem cells (BM-MSC) play important roles. BM-MSC support hematopoietic stem cells (HSC) and regulate the bone marrow immune microenvironment, but chemo/radiotherapy may damage BM-MSC and alter their functions, promoting malignant hematopoiesis.
