Summary: By using decellularized liver matrix and micropattern array chips, this study developed a system for generating size- and arrangement-controllable HepG2 spheroids for drug screening. The decellularized liver matrix showed superior biological performance in terms of cell adhesion, proliferation, and functions compared to collagen type I and Matrigel. The optimal shape and size of the micropattern were determined by evaluating the morphology, viability, and functions of the HepG2 3D cellular aggregates. Drug screening testing demonstrated that the effect of drugs on spheroids could be directly observed using confocal microscopy to measure spheroid viability.

Summary: Microbial fertilizers are commonly used as alternatives to traditional fertilizers, but the screening process for beneficial microorganisms is time-consuming. This study investigated the use of endophytic fungus-suspension cell co-culture strategy for rapid screening of plant beneficial microorganisms. Results showed that Fusarium oxysporum GU-7 significantly promoted the growth of Glycyrrhiza uralensis suspension cells and potted seedlings through its phosphorus-solubilizing ability and indole-3-acetic acid production capacity. GU-7 and GU-60 had different effects on the accumulation of secondary metabolites and altered the expression of critical genes.

This study proposes a structural optimization method that utilizes a simulated annealing algorithm to address these issues and successfully enhances the design quality of microfluidic chips through validation with test cases.
