Summary: Bioceramics, such as calcium-phosphate-based materials, are used in medical and dental implants for bone tissue replacement or repair. The effect of bioceramic sintering on intracellular signaling pathways is still unknown. This study analyzed the impact of sintering on osteoblast cell signaling pathways using sintered and non-sintered hydroxyapatite (HA) and beta-tricalcium phosphate (beta-TCP). The findings showed that sintering affects multiple signaling pathways, providing insights into bioceramic function and expanding their application scope in clinical settings.

Summary: Individual titanium and calcium-magnesium phosphates are effective sorbents with different mechanisms and efficiency depending on the phase composition. Composite materials with different Ti:(Ca+Mg) ratios were synthesized to study their sorption properties on various cations. The adsorption isotherms were analyzed using Langmuir, Freundlich, and Redlich-Peterson models. The composition of sorbents does not affect the removal of Pb2+ and Cu2+ cations, while the removal of cesium proceeds via ion exchange mechanism only. The sorption efficiency of Cd2+, Co2+, Sr2+, and Zn2+ cations on composite materials increases with the increase of titanium phosphate's share. The composite sorbents demonstrate a significant increase in the purification level compared to individual Ti and Ca-Mg phosphates.

Summary: This study compared the potential effects of three CDK4/6 inhibitors on the breast cancer bone microenvironment. The results showed that these inhibitors exerted similar inhibitory effects on the differentiation of bone resorption cells and the expression of bone resorption markers, without affecting cell viability. However, they did not affect the ability of bone formation cells to produce bone matrix, although higher doses of two inhibitors reduced the viability of these cells. In co-culture models of breast cancer and bone cells, palbociclib demonstrated a lower anti-tumor effect compared to the other two inhibitors.

Summary: This study investigated the effects of biphasic calcium phosphate (BCP) ceramics with different whiskered surfaces on macrophage polarization and functional status. The results showed that BCP ceramics with short and hollow whiskers (BCP-HW) were more conducive to protein adsorption and macrophage elongation compared to those with long and solid whiskers (BCP-W). BCP-HW ceramics down-regulated the expression of M1 macrophage markers and promoted the expression of M2 macrophage markers and growth factors. The conditioned medium derived from the co-culture of macrophages and whiskered ceramics could promote the expression of bone markers in pre-osteoblasts. In an intramuscular implantation model, BCP-HW ceramics drove the switch of macrophages to the M2 phenotype and induced ectopic bone formation.

Rspo3 deficiency impairs the inhibitory effect of Dkk1 on Wnt signaling activation, leading to the activation of Erk signaling and stabilization of beta-catenin, favoring osteoblastogenesis, bone formation, and bone mass.
