In the field of plasma medicine, cold atmospheric pressure plasma (CAP) is applied to biological matter for a therapeutic benefit. The latter is mediated by CAP-generated reactive species which treatment time-dependently either stimulate cells or induce their apoptosis. Accordingly, antitumor effects of plasma have been observed previously, but only few studies have included proper controls to compare the plasma's cytotoxicity. We here present results that murine cancer cells are more susceptible to CAP treatment compared to non-malignant fibroblasts in vitro. Additional challenge with the chemotherapeutic Gemcitabine significantly enhanced tumor cell but not fibroblast metabolic inhibition, suggesting potential combinatory effects in anticancer therapy.

Gas Plasma-Augmented Wound Healing in Animal Models and Veterinary Medicine

Sander Bekeschus, Axel Kramer, Anke Schmidt

Summary: Skin damage is inevitable and wound healing is crucial for restoring body integrity. In recent years, targeting defective wound healing has become increasingly important due to rising diseases like diabetes and vascular insufficiencies. Gas plasma technology has emerged as a promising tool for promoting wound healing by generating reactive oxygen and nitrogen species in a controlled manner.

Murine Macrophages Modulate Their Inflammatory Profile in Response to Gas Plasma-Inactivated Pancreatic Cancer Cells

Aydar Khabipov, Eric Freund, Kim Rouven Liedtke, Andre Kaeding, Janik Riese, Julia van der Linde, Stephan Kersting, Lars-Ivo Partecke, Sander Bekeschus

Summary: Gas plasma treatment of pancreatic cancer cells can alter the inflammatory profile of macrophages in a beneficial way, reducing tumor growth and metastasis without causing immunosuppression or excessive inflammation. This suggests that gas plasma therapy may be a promising approach for pancreatic cancer treatment.

Lack of Adverse Effects of Cold Physical Plasma-Treated Blood from Leukemia Patients: A Proof-of-Concept Study

Monireh Golpour, Mina Alimohammadi, Alireza Mohseni, Ehsan Zaboli, Farshad Sohbatzadeh, Sander Bekeschus, Alireza Rafiei

Summary: This study evaluated the effects of cold plasma or plasma-treated solutions on the hematologic parameters of CLL patients. The results showed that these treatments did not significantly affect the blood parameters of the patients but increased lipid peroxidation and RNS deposition, while decreasing the metabolic activity of white blood cells.

Insight into the Impact of Oxidative Stress on the Barrier Properties of Lipid Bilayer Models

Zahra Nasri, Mohsen Ahmadi, Johanna Striesow, Mehdi Ravandeh, Thomas von Woedtke, Kristian Wende

Summary: Cold physical plasma, as a new field of oxidative stress-based therapy, has the potential to generate a variety of reactive oxygen and nitrogen species (RONS), making it a promising tool for biomedical applications. However, the impact of plasma-derived RONS on cell membrane lipids and properties is not fully understood. This study investigated the changes in lipid bilayer functionality and revealed the strong impact of cholesterol on membrane permeabilization by plasma-derived species. Additionally, the chemical composition of the head group was found to influence the maintenance of barrier properties, with phosphatidylcholines being more susceptible than other lipid classes.

Periodic Exposure of Plasma-Activated Medium Alters Fibroblast Cellular Homoeostasis

Pradeep Bhartiya, Neha Kaushik, Linh N. Nguyen, Sander Bekeschus, Kai Masur, Klaus-Dieter Weltmann, Nagendra Kumar Kaushik, Eun Ha Choi

Summary: Excessive redox stress is associated with skin pathophysiologic conditions. This study explores the effect of periodic redox stress on fibroblast cells and finds that stress above a certain threshold leads to alterations in cellular processes.

Gas Plasma Protein Oxidation Increases Immunogenicity and Human Antigen-Presenting Cell Maturation and Activation

Ramona Clemen, Kevin Arlt, Thomas von Woedtke, Sander Bekeschus

Summary: Protein vaccines can induce immune responses, but inflammation caused by reactive oxygen species (ROSs) can modify proteins and affect their functionality or immunogenicity. Gas plasma technology is used to study the immunogenicity of oxidized protein antigens. The findings suggest that gas plasma-modified proteins can induce dendritic cell maturation and activation.

Heat Shock Protein 27 Affects Myeloid Cell Activation and Interaction with Prostate Cancer Cells

Debora Singer, Verena Ressel, Matthias B. Stope, Sander Bekeschus

Summary: Under oxidative stress conditions, Hsp27 is highly expressed in prostate cancer cells, playing a crucial role in anti-oxidative effects and blocking apoptosis. The presence of prostate cancer cells leads to ambiguous effects of Hsp27 on myeloid cells, altering their secretion profiles and surface marker profiles, promoting differentiation.

Oxidized Proteins Differentially Affect Maturation and Activation of Human Monocyte-Derived Cells

Ramona Clemen, Kevin Arlt, Lea Miebach, Thomas von Woedtke, Sander Bekeschus

Summary: This study investigated the effects of oxidized and native proteins on cell surface marker expression and cytokine release in human monocyte-derived cells. The results showed that oxidized proteins led to elevated expression levels of cell surface markers and modified release of cytokines. These findings are important for further exploring the application of oxidized proteins in cancer therapy.

Cold Physical Plasma-Mediated Fenretinide Prodrug Activation Confers Additive Cytotoxicity in Epithelial Cells

Mohsen Ahmadi, Debora Singer, Felix Potlitz, Zahra Nasri, Thomas von Woedtke, Andreas Link, Sander Bekeschus, Kristian Wende

Summary: Cold physical plasma is a partially ionized gas that operates at body temperature and has applications in heat-sensitive technical and medical fields. It can be used to introduce oxidative modifications to biomolecules, including the activation of anticancer prodrugs. In a proof-of-concept study, a boronic pinacol ester fenretinide prodrug was activated using an atmospheric pressure argon plasma jet. The study demonstrated that the activation of the prodrug led to enhanced cytotoxic effects in vitro, making cold physical plasma-mediated prodrug activation a promising approach for combination cancer treatment studies.
