Summary: Over the past three decades, aptamer-based biosensors have been extensively studied. This article reviews recent papers that question the binding ability of popular aptamers and provides insights into the reasons for false positive results. It also highlights the importance of carefully designed controls and immobilized libraries for obtaining high-quality aptamers. The findings emphasize the need for validated aptamers in order to achieve meaningful results in biosensor design.

Summary: This study calculated the ionization constants of sulfonylurea group antidiabetic drugs using potentiometric titration and reverse-phase liquid chromatography methods, providing valuable information for researchers studying the physicochemical properties of these drugs. The aqueous pK(a) values of the compounds were determined using Yasuda-Shedlovsky equation and mole fraction-pK(a) extrapolation methods, with results obtained for different drugs using different calculation techniques.

Summary: By characterizing the acid dissociation constants of four polyamides, the pharmacological properties of an anticancer polyamide can be optimized. The acidities of the polyamides tested were similar, while the pK(a) of the N-methylimidazole moieties varied with the peptide sequence and molecular architecture.

Summary: PLIS is an open-source software for determination of protein-ligand dissociation constants, which can be used to characterize biological processes or to shed light on biophysical aspects of interactions. In addition to analyzing experimental data, PLIS includes functionality for generating synthetic data, useful for understanding how different parameters affect the data in order to better analyze experiments.

Summary: The acid dissociation constant (pKa) of piperacillin was determined using potentiometric titration and electrospray ionization mass spectrometry (ESI-MS), which revealed two microscopic pKa values (3.37 and 8.96) corresponding to direct dissociation of the carboxylic acid group and one secondary amide group, respectively. Unlike other beta-lactam antibiotics, piperacillin undergoes direct dissociation instead of protonation dissociation. Additionally, the degradation tendency of piperacillin in alkaline solution can alter the dissociation pattern or eliminate the corresponding pKa values of amphoteric beta-lactam antibiotics. This study provides reliable determination of piperacillin's acid dissociation constant and a clear interpretation of how antibiotic stability affects the dissociation process.

Summary: The charge state of a molecule determines its interaction with the environment. Acid retention on the CelerisTM Arginine column is driven by electrostatics and charge state. The analytical retention was found to have a significant relationship with the pKa of acidic solutes, showing either a linear or quadratic trend. The developed LC mass spectrometry method is fast, reliable, accurate, easily automatized, and has broad applicability.

Summary: The commercial development of natural gas hydrates (NGH) is crucial for meeting future energy demands. Analyzing the efficiency of different production methods under various well configurations is essential for NGH exploitation. A simulation model was established based on the physical properties of hydrate reservoirs in the South China Sea. By comparing and analyzing natural gas production behavior using rapid depressurization and stepwise depressurization with a horizontal well, it was found that gravity enhances heat and mass transfer capacities in a horizontal well configuration, reducing water yield. Stepwise depressurization with a horizontal well is advantageous, as it significantly reduces water production, increases external energy input, and allows for quick reservoir heating.

Summary: Developing lime requirement (LR) methods for routine use is important in saving time, labor, and cost. This paper examined the effects of testing conditions on LR and tested a single base addition method. Optimal conditions were determined for LR estimation on Acrisols in Southeastern Vietnam. The single base addition method estimated 85.8% of LR by the reference method and can be suitable for routine use with further testing and validation.

Summary: This study extends the Kirkwood-Shumaker theory to proteins with anisotropic distributions of dissociable charges, showing that the fluctuations of the total charge and total dipole moment depend on the same proton occupancy correlator and exhibit similar dependencies on solution pH. This has significant implications for the Kirkwood-Shumaker interaction and its sensitivity to solution conditions.

Summary: In this study, it is shown that the apparent pKa measured through standard titration experiments is not sufficient to accurately assess the acidity/basicity of organic functional groups in multiprotic compounds, which is a common situation in pharmaceutical research. The use of apparent pKa in such cases may lead to costly errors. To address this issue, the authors propose pK50 as a single-proton midpoint measure, derived from a statistical thermodynamics treatment of multiprotic ionization. The study demonstrates that pK50, which can be directly measured using specialized NMR titration experiments, is superior in tracking the acidity/basicity of the functional groups across related compounds and converges to the ionization constant in the monoprotic case.

Summary: A new deep eutectic solvent (DES) composed of tetrabuthylammonium bromide (TBABr) and 1-octanol at a 1:2 molar ratio was prepared and characterized, and used as an extraction solvent for E127 in blood, urine, pharmaceutical tablet, and syrup samples. The combination of DES-UA-LLME and UV-Vis spectrophotometric detection allows for accurate determination of E127 in various sample matrices.
