Summary: Acute myeloid leukemia (AML) is a blood cancer characterized by abnormal cell proliferation and inhibition of apoptosis. The MCL-1 protein plays a critical role in the survival of AML cells. This study investigated the effects of a specific MCL-1 inhibitor, S63845, alone and in combination with other inhibitors, on AML cell lines. The results showed that S63845 decreased cell viability and induced apoptosis, and combined treatment enhanced apoptosis and induced cell differentiation. These findings suggest the potential use of MCL-1 inhibitors in combination with other pro-survival protein inhibitors.

Summary: This study aims to investigate the mechanism of MCL-1i-induced Mcl-1 protein stability. Both in vivo and in vitro experiments demonstrate that MCL-1i-induced Mcl-1 protein stability is mainly associated with defective Mcl-1 ubiquitination and concurrent apoptosis induction. Additionally, MCL-1i also enhances Mcl-1Thr163 phosphorylation via ERK pathway, contributing to Mcl-1 stability.

Summary: Multiple targeted therapies are being explored for improving clinical outcome in pediatric and young adult BCP-ALL, especially for relapsed patients. Using DBP evaluation, MEK inhibitor trametinib and multi-target tyrosine kinase inhibitor sunitinib were found to enhance apoptotic priming in NRAS-mutant and KMT2A-rearranged cell lines with high FLT3 expression, respectively.

Summary: The study reveals that upregulated lncRNA DGCR5 promotes proliferation, migration, and invasion of ESCC cells while inhibiting apoptosis. DGCR5 directly binds to SRSF1, increasing its stability and initiating important isoform switch of Mcl-1. Cell-derived xenograft model confirms DGCR5's role in facilitating ESCC tumorigenesis.

Summary: Silencing of Mcl-1 can overcome TRAIL resistance in melanoma cells and enhance the cytotoxicity of TRAIL-expressing adenovirus. The increase of apoptosis induced by Mcl-1 silencing is the main mechanism for this effect, which occurs in both TRAIL-sensitive and TRAIL-resistant melanoma cells.

Summary: The treatment of acute myeloid leukemia (AML) remains a clinical challenge, however, research shows that inhibiting the anti-apoptotic protein MCL-1 can increase apoptosis in AML cells induced by the chemotherapeutic drug Ara-C, providing a new approach for the clinical treatment of AML.

Summary: The study demonstrates that KDM4D is overexpressed in AML and can promote AML progression by regulating the expression of MCL-1, correlating with poor overall survival in AML patients. This suggests that KDM4D may serve as a novel target for the treatment of AML.

Summary: Mcl-1 protein is a significant target for cancer therapeutics, with two potential inhibitors identified in this study through screening and evaluation of drug-like compounds. These compounds showed a tendency to inhibit leukemia cell viability with Mcl-1 expressions, suggesting their potential as effective anticancer agents.

Summary: The PCSK9 enzyme interferes with LDL cholesterol metabolism. Inhibiting PCSK9 lowers LDL cholesterol levels. Monoclonal antibodies targeting PCSK9 have shown strong and persistent effects, but require frequent subcutaneous injections. Inclisiran, a synthesized siRNA, inhibits PCSK9 synthesis and provides sustained LDL cholesterol lowering with twice-yearly applications.

Summary: While mechanistic studies have shed light on the molecular mechanisms of AML and led to the development of new targeted therapies, resistance remains a significant issue in AML treatment. The exploration of circRNA in AML biology and therapy resistance shows promise in providing new insights and potential solutions to managing this disease.

Summary: Melanotic neuroectodermal tumor of infancy (MNTI) is a rare head and neck tumor that is typically diagnosed within the first year of age. Although classified as a benign tumor, some reports suggest local relapse or distant metastasis similar to a malignant tumor. This case report presents the secondary case of a 3-month-old boy with maxillary MNTI and a good prognosis after 10 years of follow-up.
