In Vitro Amoebicidal Activity of Salvia staminea and Salvia caespitosa on Acanthamoeba castellanii and Their Cytotoxic Potentials on Corneal Cells

Background/Aims: Amoebic keratitis is difficult to treat with total efficacy in some patients because of cysts, which are less susceptible than trophozoites to the usual treatments. We investigated the in vitro effectiveness of methanolic extract of Salvia staminea and Salvia caespitosa against Acanthamoeba castellanii, as well as their cytotoxicity on corneal cells in vitro. Methods: Extracts were evaluated for their amoebicidal activities using an inverted light microscope. The effect of Savia species, with concentrations ranging between 1.0 and 32.0 mg/mL, on the proliferation of A. castellanii trophozoites and cysts were examined in vitro. For determining the cytotoxicity of Salvia species on corneal cells, agar diffusion tests were performed. Results: According to the results obtained from these tests, S. staminea showed remarkable amoebicidal effect on A. castellanii. In the case of the cytotoxic activity, methanolic extract of S. staminea showed no cytotoxicity on corneal cells with a concentration of 16 mg/mL. Conclusions: Methanolic extract of S. staminea could be considered a new natural agent against Acanthamoeba. However, further evaluation by in vivo testing is needed to confirm the efficiency of its biological effect.

Summary: This study evaluated the phytochemical composition, antioxidant, and enzyme inhibitory activities of Campanula macrostachya extracts. Methanol extract showed higher antioxidant activity, while ethyl acetate and methanol extracts exhibited higher alpha-amylase and tyrosinase inhibitory activity. Flavonoids, chlorogenic acid, hesperidin, and hyperoside were found to be key compounds contributing to the observed activities.

Evaluation of the metal concentrations of wild mushroom species with their health risk assessments

Summary: This study investigated the metal contents, daily intake of metal, and health risk index values of nineteen different mushroom species collected from Uzungol, Trabzon, Turkey. The results showed that while some mushroom species are safe to consume, others exceeded the legal limits for cadmium content. Additionally, significant correlations were found between various metal pairs in the mushrooms. It was suggested that measures should be taken by the competent authorities to address potential environmental pollution in the area.

Summary: This study compared the interaction between the RBD of E484K, N501Y and South Africa triple spike variants with ACE2, and found that these mutants interacted more strongly with ACE2 than the wild-type spike. Molecular dynamics and MM-PBSA results showed that E484K and spike triple mutant complexes were more stable than the N501Y variant, with E484K and South Africa triple mutants triggering greater conformational changes in the spike glycoprotein.
