Ocular gelling microspheres: In vitro precorneal retention time and drug permeation through reconstituted corneal epithelium

Purpose: The model drug norfloxacin (NOR) was encapsulated into trehalose (TRH) and hydroxyethylcellulose (NAT) microspheres to obtain a novel gelling ophthalmic delivery system for prolonged release on corneal tissue. Methods: We assessed NOR release from microspheres, prepared by the emulsion-solvent evaporation method. A new in vitro tear turnover model, including inserts containing reconstituted human corneal epithelium (RHC), was designed to evaluate the TRH/NAT microspheres' precorneal retention time. Bioadhesive properties of TRH/NAT microspheres were validated by using drug-loaded microspheres prepared with gelatine (GLT) commonly used as reference material in adhesion studies. Results: In vitro drug release showed a typical trend of swelling systems. Precorneal retention tests showed that TRH/NAT microspheres maintained fluorescence in tear fluid for 81.7 min, whereas TRH/GLT microspheres and water solution maintained fluorescence for 51.8 and 22.3 min, respectively. NOR released from microspheres permeated throughout RHC slower (J(s) = 23.08 mu g/cm(2)h) than NOR from commercial eye drops (J(s) = 42.77 mu g/cm(2)h) used as the control. Conclusions: Adequate drug concentrations in aqueous humor could be prolonged after the administration of TRH/NAT/NOR microspheres. Good bioadhesive properties of the system and slow drug release on corneal surface might increase ocular NOR bioavailability.

Grapefruit IntegroPectin isolation via spray drying and via freeze drying: A comparison

Summary: The comparison between grapefruit IntegroPectin powders isolated via spray drying and freeze drying provides relevant information regarding phenolic content, quantification of flavonoids, radical scavenging activity, total protein content, and pH of aqueous solutions. The two drying methods yield similar pectins, except for protein content and antioxidant power. Optimizing the spray drying parameters allows for a nearly quantitative yield, approaching that obtained via freeze drying (>95%).

Chemopreventive and Anticancer Role of Resveratrol against Oral Squamous Cell Carcinoma

Summary: Oral squamous cell carcinoma (OSCC) is a prevalent and aggressive cancer, and conventional treatments have limitations. Resveratrol (RSV), a natural compound, has shown potential for treating OSCC due to its antiproliferative, antimetastatic, and proapoptotic effects. RSV also synergizes with chemotherapeutics and overcomes drug resistance. However, there is a lack of specific investigations on RSV's effects in different administration routes and vehicles. RSV-loaded liposomes have demonstrated efficacy against OSCC models, suggesting the development of drug delivery systems as a turning point in oral cancer treatment.

Summary: This study validated the potential of waste bentonite from the grape processing industries as a valuable matrix for polyphenol extraction. By applying a waste-to-market approach, functional excipients were produced. The best extract was obtained from wet bentonite with PEG200, showing the highest phenolic content and strongest antioxidant activity. It can be directly used as a polyphenol-enriched functional raw material for cosmetics and pharmaceuticals.

Buccal Thin Films as Potent Permeation Enhancers for Cytisine Transbuccal Delivery

Summary: This study aimed to design CYT-loaded buccal thin films that can promote the interaction between CYT and buccal mucosa for transbuccal drug delivery. Several compositions were prepared using the solvent casting method, and four of them showed promising aspects and flexibility. These films demonstrated homogeneity, thinness, low swelling degree, and controlled drug release, and exhibited mucoadhesive effects and potent permeation enhancement when tested on ex vivo porcine buccal mucosa.

Multicomponent Antibiofilm Lipid Nanoparticles as Novel Platform to Ameliorate Resveratrol Properties: Preliminary Outcomes on Fibroblast Proliferation and Migration

Summary: In this study, a complex mixture was designed to encapsulate Resveratrol (RSV) and produce lipid nanoparticles (LNPs) for enhanced wound healing. The optimized lipid mixture effectively encapsulated RSV and exhibited suitable nanosize and antioxidant properties. The release studies demonstrated the ability of the LNPs to release RSV under wound conditions. Biological and microbiological evaluations showed that the LNPs promoted fibroblast proliferation and migration, and exhibited antibiofilm properties at low doses. Therefore, the multicomponent LNPs could serve as a valuable RSV delivery platform for wound healing purposes.
