Kaurenoic Acid from Sphagneticola trilobata Inhibits Inflammatory Pain: Effect on Cytokine Production and Activation of the NO-Cyclic GMP-Protein Kinase G-ATP-Sensitive Potassium Channel Signaling Pathway

Kaurenoic acid [ent-kaur-16-en-19-oic acid (1)] is a diterpene present in several plants including Sphagneticola trilobata. The only documented evidence for its antinociceptive effect is that it inhibits the writhing response induced by acetic acid in mice. Therefore, the analgesic effect of 1 in different models of pain and its mechanisms in mice were investigated further. Intraperitoneal and oral treatment with 1 dose-dependently inhibited inflammatory nociception induced by acetic acid. Oral treatment with 1 also inhibited overt nociception-like behavior induced by phenyl-p-benzoquinone, complete Freund's adjuvant (CFA), and both phases of the formalin test. Compound 1 also inhibited acute carrageenin- and PGE(2)-induced and chronic CFA-induced inflammatory mechanical hyperalgesia. Mechanistically, 1 inhibited the production of the hyperalgesic cytokines TNF-alpha and IL-1 beta. Furthermore, the analgesic effect of 1 was inhibited by L-NAME, ODQ, KT5823, and glybenclamide treatment, demonstrating that such activity also depends on activation of the NO-cyclic GMP-protein kinase G-ATP-sensitive potassium channel signaling pathway, respectively. These results demonstrate that 1 exhibits an analgesic effect in a consistent manner and that its mechanisms involve the inhibition of cytokine production and activation of the NO-cyclic GMP-protein lcinase G-ATP-sensitive potassium channel signaling pathway.

Summary: The antitumor activity of inclusion complexes formed between ent-kaurenoic acid (ERKA) and beta-cyclodextrin (ERKA:beta-CD) was studied in mice, and it was found that ERKA:beta-CD300 showed significant tumor growth inhibition without side effects. This antitumor activity may be attributed to the inhibition of the NF-kB signaling pathway and LDH activity.

The nociceptive response induced by different classes of Tityus serrulatus neurotoxins: The important role of Ts5 in venom-induced nociception

Summary: This study evaluated the pain behavior induced by T. serrulatus venom and its isolated neurotoxins in mice, and found that different neurotoxins caused varying degrees of pain behavior. Additionally, the researchers developed a mice-derived polyclonal antibody targeting Ts5, which was able to neutralize the effect of this neurotoxin and significantly reduce the nociception produced by the whole venom. This study is the first to explore the effects of different classes of T. serrulatus neurotoxins on nociception, and demonstrates the important role of Ts5 in scorpion sting-induced pain.

Summary: This study evaluated whether colchicine treatment was associated with the inhibition of NLRP3 inflammasome activation in patients with COVID-19. The results showed that colchicine reduced the need for supplemental oxygen and the length of hospitalization, and lowered the serum levels of Casp1p20 and IL-18 in the first two days of treatment.

Summary: SARS-CoV-2 infection activates the NLRP3 inflammasome and leads to inflammation and severe COVID-19. Human caspase-4 (mouse caspase-11) is upregulated and activated by SARS-CoV-2, contributing to NLRP3 activation. In vivo studies using transgenic hACE2 humanized mice show that hACE2 Casp11(-/-) mice are protected from disease development, with reduced lung damage, clinical symptoms, and mortality. Analysis of human samples from fatal COVID-19 cases reveals the expression of CASP4 in patient lungs, which correlates with inflammasome components and inflammatory mediators. These findings suggest CASP4/11 as a potential target for therapeutic interventions in COVID-19.

Additionally, they can inhibit adhesion, invasion, and replication of the parasite. These compounds may serve as a new therapeutic strategy for congenital toxoplasmosis.

Synthesis and Antibacterial Activity of Polyalthic Acid Analogs

Summary: Researchers synthesized PA analogs and tested their antibacterial effects on preformed biofilms of Staphylococcus epidermidis, determining the minimal inhibitory concentration (MIC). The results showed that PA analogs 2a and 3a were able to effectively eradicate preformed biofilms of S. epidermidis and exhibited antibacterial activity against all tested Gram-positive bacteria (Enterococcus faecalis, Enterococcus faecium, S. epidermidis, Staphylococcus aureus) with varying MIC values. Therefore, further in vitro and in vivo testing is warranted to develop antibiotic formulations targeting Gram-positive bacteria.

Polyalthic Acid from Copaifera lucens Demonstrates Anticariogenic and Antiparasitic Properties for Safe Use

Summary: This study evaluated the potential of Copaifera lucens in combating caries and toxoplasmosis. The results showed that the oleoresin and compound from Copaifera lucens were effective in inhibiting cariogenic bacteria and hindering the intracellular proliferation of Toxoplasma gondii. The study also found that the Copaifera lucens extract exhibited low toxicity. These findings highlight the potential of Copaifera lucens as alternative treatments for caries and toxoplasmosis.

Oxidative activity of corpus luteum and ovarian parenchyma in Bos taurus indicus heifers

Summary: The aim of this study was to evaluate oxidative stress in ovaries and corpus luteum (CL) of Bos taurus indicus females and the oxidant effect of CL in ovarian tissues. The results showed that luteal tissues had higher antioxidant activity than ovarian tissues, and there was no difference in antioxidant activity and oxidative stress between ovarian fragments obtained from different regions (with or without CL). The presence of CL in the ovaries did not influence oxidative stress or antioxidant potential.

Pre-emergence herbicidal efficiency and uptake of atrazine-loaded zein nanoparticles: a sustainable alternative to weed control

Summary: In this study, zein nanoparticles associated with the herbicide atrazine were developed and their efficacy in weed control was evaluated. The nanoformulation showed good stability and encapsulation efficiency. It exhibited greater effectiveness against the target plant, Brassica juncea, even at low doses, without causing toxicity to the crop species. The nanoparticles were captured and accumulated in roots but had limited transportation to the shoot. The atrazine nanoencapsulation did not increase the mobility of the herbicide in the soil.
