Summary: This study found that microsatellite alterations are common in both mouse and human induced pluripotent stem cells, but identified a type of human iPSC with significantly reduced alterations. By generating reprogrammed stem cell lines with the same genetic background, somatic de novo mutations could be comprehensively detected and validated, showcasing the effectiveness of using sister clones. This approach was successfully applied to human iPSCs, providing a significant advancement in addressing genome abnormalities in iPSCs.

Summary: Genetic mapping is crucial in narrowing down candidate mutations and genes corresponding to mutant phenotypes. This study introduces a mapping toolkit for use in Caenorhabditis elegans, utilizing insertion-deletion polymorphisms for efficient and cost-effective mapping. The method minimizes the need for large populations of mutants and showcases its strengths and weaknesses through mapping of known and unknown mutations.

Summary: Atovaquone-proguanil is commonly prescribed for malaria prophylaxis, but sporadic mutations conferring resistance have been detected in recent years. Monitoring genetic polymorphisms associated with drug resistance is crucial for assessing prevalence and designing control strategies. This study developed primers and validated a high-throughput Ligase detection reaction fluorescent microsphere assay (LDR-FMA) to detect SNPs associated with atovaquone resistance in clinical samples. The results were consistent with DNA sequence data, demonstrating the potential of this method as a tool for detecting genetic polymorphisms in P. falciparum.

Summary: The DHPV-U method successfully detected DHPV in both M. rosenbergii and P. monodon, with a high sequence identity. The findings support the hypothesis that the HP virus in M. rosenbergii is also a hepanhamaparvovirus.

Summary: Almost every animal trait is strongly associated with parasitic infection or potential exposure to parasites. Accurately determining the status and severity of endoparasitic infection without killing and dissecting the host is a major challenge researchers face. This study demonstrates a noninvasive and minimally stressful method for detecting endoparasitic infection with greater accuracy than dissection alone, improving protocols for live animal studies.

Summary: The study focuses on using a PCR-based method to detect a single copy T-DNA insertion in Arabidopsis transformants. Analysis of the gray ratio between two PCR products allows for determination of whether the test transformants contain a single copy of T-DNA insertion, with reference to control lines for quantitative analysis.

Summary: This study proposed a model combining 3D fluorescence spectra with wavelet moments for identifying adulterated sesame oil, which effectively reduced noise and data volume through wavelet multiresolution decomposition. The extracted features were validated by hierarchical clustering, and target wavelet moments for identifying adulterated sesame oil were determined, providing a fast and expandable method for online measurement and reference for vegetable oil authentication.

Summary: A new triplex real-time PCR method was established in this study for the specific identification of ASFV wild-type and gene deletion strains, preventing non-specific amplification of other swine viruses. Of the 1646 samples analyzed, 67 were positive for ASFV, all corresponding to the wild-type virus. The minimum copy number of standard plasmid DNA detected using the method was determined for B646L, EGFP, and mCherry genes.

Summary: Shortly after its emergence, Omicron and its sub-variants quickly replaced the Delta variant during the current COVID-19 outbreaks. A robust and economical real-time PCR method is needed to detect and identify multiple different circulating variants for surveillance and diagnostic applications.

Summary: This paper presents a simple alternative to the delta method for calculating confidence intervals of nonlinear functions of model parameters. The proposed method performs better when the estimates of model parameters have an approximate multivariate normal distribution, and a diagnostic tool is provided to assess the reliability of this assumption. Compared to other methods, it is computationally efficient and provides better coverage properties without the need to refit the model.

Summary: Entomopathogenic fungi play an important role in mosquito control. A simple method to detect fungi on living mosquito larvae exposed to fungal pathogens was tested in this study. The technique facilitates the detection and isolation of entomopathogenic fungi, providing a compact, convenient, and rapid means to survey larval mosquito populations for fungal pathogens in the field.
