Abstract

The prevalence of diabetes throughout the world continues to increase and has become a major health issue. Recently there have been several reports of inhibitors directed toward the sodium-dependent glucose cotransporter 2 (SGLT2) as a method of maintaining glucose homeostasis in diabetic patients. Herein we report the discovery of the novel O-xyloside 7c that inhibits SGLT2 in vitro and urinary glucose reabsorption in vivo.

Cardiovascular Diseases: Therapeutic Potential of SGLT-2 Inhibitors

Summary: Cardiovascular diseases (CVD) are a major global health concern, and sodium/glucose cotransporter 2 (SGLT2) inhibitors have emerged as a promising class of medications for managing CVD. In this paper, we discuss the role of various SGLT-2 inhibitors as potential therapeutic targets for cardiovascular diseases, highlighting their significant cardiovascular benefits and renal protection. These novel therapeutic approaches aim to improve patient outcomes and reduce the burden of these conditions, but further studies are needed to explore the scientific advancements and mechanisms.

From Kidney Protection to Stroke Prevention: The Potential Role of Sodium Glucose Cotransporter-2 Inhibitors

Summary: Chronic kidney disease (CKD) is a significant risk factor for stroke and cerebrovascular disease, yet the interaction between the brain and kidneys has received less attention compared to the interaction between the heart and kidneys. This article focuses on the potential of SGLT2 inhibitors, a class of oral anti-diabetic drugs, in preventing strokes and improving the cerebro-renal interaction, regardless of diabetes status. The authors propose the roles of baseline renal functions and dual inhibition of SGLT1/2 in stroke prevention, as well as the additional benefits of reducing atrial fibrillation and hemorrhagic stroke.

The effects of Sodium-glucose cotransporter 2 inhibitors on adipose tissue in patients with type 2 diabetes: A meta-analysis of randomized controlled trials

Summary: This study systematically evaluated the effect of SGLT2 inhibitors on adipose tissue in patients with type 2 diabetes. Meta-analysis results showed that the use of these inhibitors significantly reduced visceral adipose tissue, subcutaneous adipose tissue, body weight, and triglycerides in type 2 diabetes patients, which may be attributed to their protective effect on the cardiovascular system.

Summary: This study investigated the affinity of alpha-methyl-4-deoxy-4-[F-18]fluoro-d-glucopyranoside (Me4FDG) for SGLT1 and SGLT2 proteins, and its role in predicting response to SGLT2 inhibitors in patients with type 2 diabetes. The results showed that Me4FDG excretion is a reliable predictor of the therapeutic effectiveness of SGLT2 inhibitors.

The safety of sotagliflozin in the therapy of diabetes mellitus type 1 and type 2: A meta-analysis of randomized trials

Summary: This meta-analysis demonstrates that sotagliflozin has an acceptable safety profile in patients with diabetes, with adverse events including genital mycotic infection, related-to-acidosis events, diarrhea, volume depletion, and severe nocturnal hypoglycemia events. The subgroup analysis of sotagliflozin dosage is of great clinical significance for guiding the use of sotagliflozin in patients with diabetes in the future.

Poor adherence and persistence to sodium glucose co-transporter 2 inhibitors in real-world settings: Evidence from a systematic review and meta-analysis

The Effects of SGLT2 Inhibitors on Liver Cirrhosis Patients with Refractory Ascites: A Literature Review

Summary: Decompensated liver cirrhosis often leads to refractory ascites, which is a poor prognostic factor for patients with liver cirrhosis. Current treatment for ascites in cirrhosis patients involves the use of aldosterone blockers, loop diuretics, and sometimes vasopressin receptor antagonists. Recent reports suggest that sodium-glucose cotransporter 2 (SGLT2) inhibitors may provide a new treatment option with different mechanisms for refractory ascites. The mechanisms of ascites reduction with SGLT2 inhibitors include natriuresis, osmotic diuresis, improvements in glucose metabolism and nutritional status, hepatoprotection, amelioration of the sympathetic nervous system, and inhibition of the renin-angiotensin-aldosterone system. This literature review discusses previous cases in which SGLT2 inhibitors effectively treated ascites caused by liver cirrhosis and provides insights into the potential use of SGLT2 therapy for ascites in the future.

Summary: Abnormality in glucose homeostasis due to hyperglycemia or insulin resistance is the hallmark of type 2 diabetes mellitus, leading to cellular dysfunction and diabetic cardiomyopathy. New antihyperglycemic agents such as GLP-1 agonists and SGLT2 inhibitors have shown cardioprotective effects by attenuating endothelial dysfunction and improving cardiovascular safety. The mechanism behind these effects is not fully understood, but there is evidence suggesting an interaction between NHE and SGLT2i in cardiovascular homeostasis.
