Structure-activity relationship studies for the peptide portion of the bladder epithelial cell antiproliferative factor from interstitial cystitis patients

We performed comprehensive structure-activity relationship (SAR) studies on the peptide portion of antiproliferative factor (APF), a sialylated frizzled-8 related glycopeptide that inhibits normal bladder epithelia] and urothelial carcinoma cell proliferation. Glycopeptide derivatives were synthesized by solid-phase methods using standard Fmoc chemistry and purified by RP-HPLC; all intermediate and final products were verified by HPLC-MS and NMR analyses. Antiproliferative activity of each derivative was determined by inhibition of (3)H-thymidine incorporation in primary normal human bladder epithelial cells. Structural components of the peptide segment of APF that proved to be important for biological activity included the presence of at least eight of the nine N-terminal amino acids, a negative charge in the C-terminal amino acid, a free amino group at the N-terminus, maintenance of a specific amino acid sequence in the C-terminal tail, and trans conformation for the peptide bonds. These data provide critical guidelines for optimization of structure in design of APF analogues as potential therapeutic agents.

Summary: In the URRAH study, we identified the cut-off value for the ratio of serum uric acid (SUA) to serum creatinine (sCr) that can predict cardiovascular events. We found that having SUA/sCr > 5.35 is an independent cardiovascular risk indicator for both men and women.

Summary: This study investigates the association between very high HDL-cholesterol levels and cardiovascular mortality (CVM), and whether this association is influenced by hyperuricemia. The results show a J-shaped association between high HDL-cholesterol levels and CVM in women, but not in men. Additionally, the risk of CVM related to high HDL-cholesterol is increased in women with hyperuricemia.

Summary: A wide range of glycan structures on cell surfaces have significant roles in various biological processes and immune functions. Immune responses to foreign carbohydrate antigens and tumour-associated carbohydrate antigens are important for microbial clearance and anti-tumour vaccines. The linkage point of antigenic glycans affects their presentation to the immune system as well as carbohydrate binding molecules. The understanding of these presentations provides insights into the design of vaccines and recognition mechanisms.

Summary: High serum uric acid (SUA) levels have been found to predict all-cause mortality (ACM) and cardiovascular mortality (CVM) in cardiometabolic patients without established cardiovascular disease, regardless of triglyceride (TG) levels. The study included 8124 patients from the URic acid Right for heArt Health (URRAH) study cohort, and the findings were confirmed by exploratory and sensitivity analyses. This suggests that SUA plays a substantial role in predicting cardiovascular risk in both normotriglyceridemia and hypertriglyceridemia.

Summary: This study demonstrates a significant association between NDUFC2 variants and left ventricular hypertrophy (LVH) in hypertensive patients. NDUFC2 variants result in reduced gene expression and impaired mitochondrial function, which contribute to the development of cardiac hypertrophy. In vitro experiments show that Ndufc2 deficiency leads to mitochondrial dysfunction and cardiomyocyte hypertrophy.

Low Perception of Obesity as a Pathological Condition Among Italian Cardiologists

Summary: This study investigated the perception and importance of treating obesity among 137 cardiologists and residents in cardiology. The results showed that most of the participants underestimated the significance of obesity and rarely measured waist circumference and waist-to-hip ratio in their clinical practice. They also showed reluctance to prescribe anti-obesity drugs. This highlights a lack of awareness and importance given to obesity as a chronic disease and a modifiable cardiovascular risk factor.
