method for the simultaneous determination of the cardiotonics dopamine and 4-methyl-2-aminopyridine in serum, previously derivatized with fluorescamine, by high performance liquid chromatography coupled with a fluorescence detector is presented. Chromatographic and detection conditions were optimized; a LiChrospher 100 RP-18 reversed phase column (250 x 4 mm; 10 mu m) and the mobile phase of methanol: water, in gradient mode, were found as the most suitable. The derivatives were eluted in 10 min with good reproducibility at a flow rate of 0.9 mL/min, being 20 mu L the volume of sample injected. The detection was done at 490 nm using borax buffer solution (pH 8) for both cardiotonics. Relative standard deviations (n = 10) of 6.7% and 5.9% (concentration) and 2.4% and 3.05% (retention time), were obtained for dopamine and 4-methyl-2-aminopyridine, respectively. Recoveries of the spiked female rabbit serum ranged from 90% to 110%, and detection limits of 122 ng/mL for dopamine and 108 ng/mL for 4-methyl-2-aminopyridine, were obtained.

Detection of Ru potential metallodrug in human urine by MALDI-TOF mass spectrometry: Validation and options to enhance the sensitivity

Nadia Nunes, Iva Popovic, Elder Abreu, Dina Maciel, Joao Rodrigues, Juan Soto, Manuel Algarra, Marijana Petkovic

Summary: The detection of RuCp complex in human urine using MALDI-TOF MS without prior purification or removal of inorganic salts was successfully achieved. Different acquisition conditions showed varying levels of sensitivity and precision, with the matrix-free approach and DHB matrix demonstrating the highest sensitivity and precision in urine samples. Higher sensitivity of matrix-free MALDI detection of RuCp can be achieved by adding carbon dots to the urine solution.

Nitrene formation is the first step of the thermal and photochemical decomposition reactions of organic azides

Juan Soto, Manuel Algarra, Daniel Pelaez

Summary: In this study, the decomposition of isopropyl azide in both ground and excited states was investigated using multiconfigurational CASSCF and MS-CASPT2 electronic structure approaches. The results revealed a stepwise mechanism for the thermal decomposition of azide, with the formation of isopropyl nitrene being the rate-determining step. Two routes were identified for the decomposition, and the associated intermediates were characterized. Kinetic modeling showed that the singlet channel had significantly higher rate coefficients than the spin-forbidden channel. Furthermore, the concerted mechanism for the formation of the imine derivative was ruled out after comparing different calculation methods.

CASPT2 study of the electronic structure and photochemistry of protonated N-nitrosodimethylamine (NDMA-H+) at 453 nm

Juan Soto, Daniel Pelaez, Manuel Algarra

Summary: In this study, the photodissociation of protonated derivatives of N-nitrosodimethylamine was investigated using the CASPT2 method. It was found that only one of the possible protonated species, N-nitrosoammonium ion, absorbed in the visible region and dissociated to produce aminium radical cation and nitric oxide. The intramolecular proton migration reaction was found to be inaccessible in both the ground and excited state. In acidic solutions of aprotic solvents, only the formation of [(CH3)(2)NH-NO](+) was observed.
