Complement-Fixing Anti-Type VII Collagen Antibodies Are Induced in Th1-Polarized Lymph Nodes of Epidermolysis Bullosa Acquisita-Susceptible Mice

The environment encountered in secondary lymphoid organs (e.g., lymph nodes) influences the outcome of immune responses. Immunization of mice with type VII collagen, an adhesion protein expressed at the cutaneous basement membrane, induces experimental epidermolysis bullosa acquisita (EBA). In this model, clinical disease is associated with the H2s haplotype of the MHC found in SJL/J mice. Most other strains (e.g., BALB/c, C57BL/6, NZM2410/J) are resistant to clinical disease, despite autoantibody production. Comparison of autoantibody response in EBA-resistant and -susceptible mice showed an IgG2-dominated response in the latter. We hypothesized that EBA susceptibility is due to specific cytokine gene expression in draining lymph nodes (dLN). To challenge this hypothesis, EBA-susceptible (SJL/J) and -resistant (BALB/c, C57BL/6) mice were immunized with type VII collagen, followed by analysis of clinical phenotype, subclasses of circulating and tissue-bound autoantibodies, complement activation, and cytokine gene expression in dLN. Disease manifestation was associated with induction of complement-fixing autoantibodies, confirming previous observations. Furthermore, however, IFN-gamma/IL-4 ratio in dLN of EBA-susceptible mice was significantly increased compared with EBA-resistant strains, suggesting a Th1 polarization. Immunization of H2s-congenic C57BL/6 mice (B6. SJL-H2s) led to Th1 polarization in dLN and clinical disease. In addition to their cytokine milieu, EBA-susceptible and -resistant mice also differed regarding the expression of Fc gamma R on peripheral leukocytes, in which a higher Fc gamma RIV expression in SJL/J and B6. SJL-H2s mice, compared with C57BL/6, was associated with skin lesions. In summary, blistering in experimental EBA is regulated by both adaptive (divergent class switch recombination due to polarized cytokine expression) and innate (Fc gamma R expression) immune mechanisms. Association of Rituximab With Risk of Long-term Cardiovascular and Metabolic Outcomes in Patients With Pemphigus

Risk factors and sequelae of epidermolysis bullosa acquisita: A propensity-matched global study in 1,344 patients

Summary: Identification of risk factors and sequelae of diseases is crucial for primary prevention and disease management. This study used TriNetX to identify risk factors and sequelae of the rare autoimmune disease EBA. The findings revealed chronic inflammatory diseases, especially lupus erythematosus and lichen planus, as the highest risk factors for EBA development and metabolic and cardiovascular diseases, and thrombosis as the most common sequelae after EBA diagnosis.

Impact of diet and host genetics on the murine intestinal mycobiome

Summary: In this study, 591 mice were used to demonstrate that fungi are regulated by host genetics and that diet has a regulatory role in the composition of gut fungi. The role of fungi in complex interactions involving host genetics, bacteria, and environmental factors remains understudied. Using whole genome sequencing and genotyping, quantitative trait loci associated with various fungal species were mapped in mice. Additionally, the study identified fungal indicator species associated with different dietary regimes.

EGFR inhibition leads to enhanced desmosome assembly and cardiomyocyte cohesion via ROCK activation

Summary: Arrhythmogenic cardiomyopathy (AC) is a familial heart disease caused by impaired desmosome turnover, and stabilizing desmosome integrity through EGFR and ROCK inhibition may offer new treatment options. In this study, EGFR inhibition enhanced cardiomyocyte cohesion and increased DSG2 recruitment and binding at cell borders. Additionally, EGFR inhibition led to the upregulation of ROCK, and ROCK inhibition abolished the effects of EGFR inhibition on desmosome assembly and cardiomyocyte cohesion. These findings suggest that targeting EGFR and ROCK could be potential therapeutic strategies for AC.

Pruritus Is Associated with an Increased Risk for the Diagnosis of Autoimmune Skin Blistering Diseases: A Propensity-Matched Global Study

Summary: Autoimmune bullous skin diseases (AIBDs), such as bullous pemphigoid (BP) and pemphigus, are characterized by autoantibodies targeting structural proteins. Pruritus is common in patients with BP and pemphigus, and can be the sole symptom in some cases. The temporal relationship between pruritus and AIBDs is unclear, and the presence of pruritus in other AIBDs is also unknown.

LS presents as chronic whitish atrophic patches with itching and soreness in the genital and anal regions. Treatment options include topical corticosteroids and calcineurin inhibitors.
