Summary: The study demonstrated that S100A4 plays a critical role in regulating mast cell activation and allergic responses. S100A4(-/-) mice showed reduced symptoms in allergic models, indicating that S100A4 is required for mast cell activation and may be involved in the regulation of allergic diseases through this mechanism.

Summary: The study found that mimicking allergic lung inflammation in mice can lead to significant histological changes in the lungs and behavioral changes consistent with depression and altered spatial memory. The increased levels of certain proteins suggest a link between lung inflammation and brain mast cell activation, which may be responsible for depression-like behavior.

Summary: Mast cells are widely distributed immune cells with diverse phenotypes and functions. They exhibit high functional heterogeneity and plasticity due to their unique maturation mechanism. Mast cells can adapt to different microenvironments and regulate allergy and inflammation under the peripheral immune education system. This review focuses on the mechanism and heterogeneity of mast cell migration to mucosal tissues, such as the gastrointestinal tract. The immunosuppressive properties of mast cells suggest a potential new approach for treating inflammatory and allergic disorders.

Summary: The study demonstrated that ursolic acid (UA) exerts anti-allergic effects by inhibiting FceRI-mediated mast cell activation. It suppresses mast cell degranulation by reducing intracellular calcium levels and inhibits the expression and release of inflammatory cytokines. Furthermore, UA also inhibits passive cutaneous anaphylaxis and active systemic anaphylaxis reactions in a mouse model in a dose-dependent manner.

Summary: In this study, the natural isoflavone formononetin (FNT) was found to inhibit IgE-mediated mast cell activation and reduce allergic inflammation. FNT suppressed the release of inflammatory factors, histamine, and beta-hexosaminidase, and inhibited NF-kappa B and MAPK activity in mast cells. FNT also induced degradation and ubiquitination of the high-affinity IgE receptor, suggesting its potential as a therapeutic target for allergic diseases.

Summary: Acute lung injury (ALI) is a common and devastating clinical disorder with a high mortality rate and no specific therapy. The aim of this study was to evaluate the protective effects of Adelmidrol (ADM) in a mice model of ALI induced by lipopolysaccharide (LPS). ADM administration reduced lung damage, airway infiltration, and oxidative stress by regulating mast cells and antioxidant enzymes. These findings suggest that ADM could be a potential candidate in the management of ALI.

Summary: This study investigates the pro-resolution role of mast cells in allergic inflammation. The results show that activated mast cells release a significant amount of resolvin D1 after 30 minutes, which can actively contribute to the resolution of allergic inflammation.

Summary: ILC2s resident in the lung are activated by IL-33 and can migrate to peripheral blood and liver, leading to eosinophilic inflammation and liver damage. Nasal IL-33 pre-treatment attenuates allergic inflammation and liver diseases.

Summary: This study elucidated the non-transcriptional role of IRF3 in mast cell activation and allergic inflammation. In vitro and in vivo experiments showed that IRF3 played a significant role in mast cell-mediated allergic inflammation. The activation of IRF3 was observed during mast cell activation process, and it affected the production of granule contents and the anaphylaxis responses. Overall, IRF3 is an important factor inducing mast cell activation and regulating HDC activity.

Summary: A mast cell-dependent murine model of allergic peritonitis was established by inducing inflammation using ovalbumin and Staphylococcus aureus enterotoxin B. The peak of peritoneal inflammation was observed 48 hours after challenge, followed by a decrease starting at 96 hours and complete clearance by 168 hours. Additionally, higher levels of tryptase and increased vascular permeability were found 30 minutes post-challenge.

Summary: Metabolic programming is closely linked to immune cell function, with mTOR signaling as a key regulator. While mTORC2 signaling is crucial for M2 macrophage generation, the role of mTORC1 signaling remains controversial. Inhibiting mTORC1 signaling enhances M1 macrophage function, despite defects in glycolytic metabolism, by inhibiting sirtuins and enhancing histone acetylation.
