A fluorescent probe CPO was developed to detect SO2 derivatives in body and food, showing high energy transfer efficiency and selectivity. It had fast response time and low detection limit, and could detect SO2 derivatives in various substances.

Summary: Intracellular pH plays a crucial role in cellular processes, and a water-soluble fluorescent pH probe based on the protonation/deprotonation of the 4-methylpiperazin-1-yl group was designed. The probe showed fluorescence OFF-ON mechanism under acidic conditions, exhibiting high selectivity, photostability, fast response to pH changes, and low cytotoxicity to cells. It can selectively accumulate in lysosomes and monitor pH changes stimulated by chloroquine in living cells. The probe has potential for diagnosing pH-related diseases.

Summary: In this study, a novel FRET-based fluorescent probe (named CBC) was developed for highly selective detection of bisulfite using a ratiometric method. Probe CBC showed excellent sensitivity towards HSO3- within 1 equivalent. Cell imaging experiments demonstrated that CBC could detect exogenous and endogenous bisulfite with excellent photostability and low cytotoxicity. Moreover, CBC probe achieved specific targeting to lipid droplets, indicating its potential as a biological tool for monitoring in vivo bisulfite changes.

Summary: Viscosity and pH are crucial microenvironment parameters that can indicate various diseases when experiencing abnormal fluctuations at the cellular level. Traditional viscosity/pH meters are not suitable for cellular environments, so there is an urgent demand for developing viscosity/pH-sensitive fluorescent probes. The novel dual-functional camphor-based fluorescent probe PN-SP exhibits intense green emission in response to alkaline pH and bright red fluorescence in high-viscosity media, offering a solution to problems caused by the use of multiple probes in cellular environments.

It provides valuable insights for the design and development of new smart and flexible-functionality ROMP-based polymeric materials for various biological applications.

Summary: Myogenesis is regulated by MYOD1 and involves ADAMTSL2, which acts as a signal hub to integrate WNT, TGF6, and potentially other signaling pathways in the dynamic microenvironment of differentiating myoblasts during skeletal muscle development and regeneration. ADAMTSL2 depletion leads to impaired myoblast differentiation and abnormal skeletal muscle architecture. Mechanistically, ADAMTSL2 enhances WNT signaling by binding to WNT ligands and receptors, and a WNT-binding ADAMTSL2 peptide is sufficient to promote myogenesis in vitro.

Summary: Biochemical and biophysical factors are important to consider when modeling in vivo cellular behavior in vitro. One underappreciated factor is the high concentration of macromolecules present in vivo, which is typically not simulated in standard cell culture conditions. This study found that adding inert macromolecules to cell culture medium can better mimic the high concentration environments found in vivo, leading to enhanced deposition of fibrillin-1.
