Summary: Hypeptin is a cyclodepsipeptide antibiotic that blocks bacterial cell wall biosynthesis by binding to multiple biosynthesis intermediates, forming a 2:1 complex. It exhibits potent activity against gram-positive pathogens and resistance to it does not readily develop. Detailed analysis of the hypeptin biosynthetic gene cluster supports a model for the synthesis of the octapeptide.

Summary: Antibiotic resistance is a major concern, and new antibiotics with novel chemistry and mechanisms of action are needed. The recently discovered antibiotic cacaoidin is the first class V lanthipeptide, combining unique structural features and D-amino acid content. It shows antimicrobial activity against gram-positive pathogens and interferes with peptidoglycan biosynthesis. Further studies have demonstrated that cacaoidin exhibits a dual mode of action by binding to lipid IIPGN and inhibiting cell wall transglycosylases.
