Beneficial Effects of Donepezil on Vascular Endothelial Dysfunction-Associated Dementia Induced by L-Methionine in Rats

Donepezil (acetyl cholinesterase inhibitor) is a mainstay of clinical intervention to contain memory deficits of Alzheimer's disease. However, its beneficial role in endothelial dysfunction-associated dementia i.e. vascular dementia still needs to be explored. The present study was designed to investigate the effect of donepezil on vascular endothelial dysfunction. and associated memory deficits in rats. Atorvastatin (3-hydroxy-4-methyl-glutaryl (HMG)-CoA inhibitor) was taken as the standard. Rats were administered L-methionine (1.7 g/kg per day, p.o., 4 weeks and 4 days i.e. chronic treatment) to produce endothelial dysfunction and dementia. Serum nitrite level was estimated as a marker of endothelial function. Morris water maze (MWM) test was employed for assessment of memory. Brain tissue thiobarbituric acid reactive substances (TBARS) and reduced glutathione (GSH) were estimated to assess oxidative stress. Brain acetylcholine esterase (AChE) activity and serum total cholesterol level were also estimated. L-methionine produced endothelial dysfunction as reflected by significant decrease of serum nitrite concentration. L-methionine-treated rats performed poorly on MWM indicating impairment of memory as well. These rats also showed a significant rise in brain oxidative stress, AChE activity and serum total cholesterol levels. Donepezil (0.1 mg/kg p.o.) and atorvastatin (10 mg/kg p.o.) attenuated L-methionine-induced endothelial dysfunction. This intervention reversed L-methionine-induced rise of brain oxidative stress and AChE activity. Furthermore, atorvastatin produced a reduction of L-methionine-induced rise in serum cholesterol. The beneficial effects of donepezil may be attributed to its multiple effects and this study highlights the potential of donepezil in vascular dementia.

Attenuation of neurobehavioural abnormalities by papaverine in prenatal valproic acid rat model of ASD

Summary: This study investigated the effects of a PDE10A inhibitor, papaverine, on core phenotypes of ASD in a Pre-VPA model in rats. Papaverine administration improved social behavior, anxiety, locomotion, and nociception, while also impacting protein markers associated with neuronal function. The findings suggest that targeting PDE10A could be a potential strategy for further understanding ASD phenotypes.

Papaverine ameliorates prenatal alcohol-induced experimental attention deficit hyperactivity disorder by regulating neuronal function, inflammation, and oxidative stress

Summary: ADHD is a neurodevelopmental disorder with complex etiology, and PDE10A has been implicated as a potential target for further understanding its phenotypes. Administration of the PDE10A inhibitor papaverine improved hyperactivity, inattention, and anxiety in an ADHD model, possibly by altering protein markers associated with neuronal function and brain inflammation.

Various nanotechnology-based drug delivery systems, such as solid lipid nanoparticulate drug delivery systems and emulsion-based nanodrug delivery systems, play crucial roles in the targeted delivery of poorly soluble drugs. This review provides a comprehensive overview of lipid nanoparticles as carriers of bioactive molecules, covering aspects like synthesis, characterization, advantages, disadvantages, toxicity, and applications in the medical field. Recent developments in patents and clinical trials related to solid lipid nanoparticles (SLNs) and nanostructure lipid carriers (NLCs) are also discussed.

Summary: An Ultra-High-Performance Liquid Chromatography (UHPLC) method was developed and validated for the analysis of 16 PAH compounds in soil samples from Manesar, India. The study found that PAH concentrations were highest in the winter season in industrial areas, while the lowest levels were observed during the monsoon season. The PAH levels in Manesar were relatively lower compared to Brazil, UK, Germany, and Korea.

Protective Effect of Nimodipine Against Valproic-acid Induced Biochemical and Behavioral Phenotypes of Autism

Data on antibiotic resistance among indoor microbiome at Meerut, India

Summary: This study surveyed the microbial community and their antibiotic profiling in the domestic environment of rural and urban areas in Meerut city, India. The results revealed a higher bacterial load compared to fungal load, with kitchens being more contaminated than living rooms. The dominant bacteria were Pseudomonas sps, Citrobacter sps, Bacillus Subtilis, Brevundimonas diminuta, Bacillus megaterium, and Klebsiella pneumoniae. Antibiotic resistance was mainly derived from plasmids.
