Antitumor and immunomodulatory activity of polysaccharides from the roots of Actinidia eriantha

Aim of the study: The roots of Actinidia eriantha Benth (Actinidiaceae) have been used for cancers in the Chinese folk medicine. The present study aimed at evaluating the antitumor potentials of the polysaccharides from the roots of Actinidia eriantha and elucidating their immunological mechanisms by determining the effects on the growth of tumor transplanted in mice and the immune response in tumor-bearing mice. Materials and methods: The total polysaccharide AEP and fours purified polysaccharides AEPA, AEPB, AEPC and AEPD were isolated and purified from the roots of Actinidia eriantha by hot water extraction, ethanol precipitation, dialysis and gel filtration. Their effects on the growth of mouse transplantable tumor, splenocyte proliferation, the activity of natural killer (NK) cells and cytotoxic T lymphocytes (CTL), production of cytokines from splenocytes, and serum antigen-specific antibody levels in tumor-bearing mice were measured. Results: AEP and four purified polysaccharides could not only significantly inhibit the growth of mouse transplantable tumor, but also remarkably promote splenocytes proliferation, NK cell and CTL activity, IL-2 and IFN-gamma production from splenocytes, and serum antigen-specific antibody levels in tumor-bearing mice. Conclusions: The antitumor activity of AEP and four purified polysaccharides might be achieved by improving immune response, and the composition of the monosaccharides, uronic acid contents and molecular weight could affect their antitumor and immunomodulatory activity. Microplastics aggravate the bioaccumulation of three veterinary antibiotics in the thick shell mussel Mytilus coruscus and induce synergistic immunotoxic effects

Summary: The study revealed that exposure of mussels to antibiotics and microplastics led to synergistic immunotoxic effects, including significant reductions in phagocytic rate and total hemocyte count, increases in intracellular reactive oxygen species and decreases in F-actin cytoskeleton, cell viability, and immune-related gene expression. Coexposure to antibiotic-MP also resulted in higher accumulation of antibiotics, possibly explaining the observed synergistic immunotoxic effect. Additionally, exposure to pollutants suppressed glutathione-S-transferase activity, downregulated detoxification-related genes, and induced lipid peroxidation in gills, potentially facilitating the entry of antibiotics and increasing their accumulation.

Caspase-1-Dependent Pyroptosis Mediates Adjuvant Activity of Platycodin D as an Adjuvant for Intramuscular Vaccines

Summary: This study identified the potential signaling pathways involved in the adjuvant activity of Platycodin D (PD) using in vitro and in vivo models. The results showed that PD induced cytotoxicity and inflammatory response via the caspase-1-dependent NLRP3 inflammasome signaling pathway in C2C12 cells. In vivo analysis further demonstrated that blocking NLRP3 and caspase-1 inhibited PD-induced cytokine production and immune cell recruitment, and impaired the adjuvant activity of PD in mice. These findings expand our understanding of the mechanisms of action of saponin-based adjuvants.

Adjuvant effect of two polysaccharides from the petals of Crocus sativus and its mechanisms

The results showed that these polysaccharides enhanced immune responses, triggered the production of cytokines, and facilitated antigen transport, making them promising vaccine adjuvant candidates.

Herba Origani alleviated DSS-induced ulcerative colitis in mice through remolding gut microbiota to regulate bile acid and short-chain fatty acid metabolisms

Summary: This study investigated the protective effect of Herba Origani on dextran sodium sulfate (DSS)-induced ulcerative colitis in mice. The results showed that Herba Origani alleviated colitis symptoms, inflammation, and pathological injury, and repaired intestinal barrier function. It also restored gut microbiota dysbiosis, regulated bile acids metabolism, and adjusted short-chain fatty acids (SCFAs). These findings suggest that Herba Origani has the potential to be used for the treatment of inflammatory intestinal diseases.

Adjuvant activity of tubeimosides by mediating the local immune microenvironment

The TBMs significantly boosted the immune responses and influenced the expression of immune, chemotaxis, and inflammation-related genes. The interaction between TBM I and SYK and LYN was found to be involved in its adjuvant activity.

Isorhamnetin as a novel inhibitor of pneumolysin against Streptococcus pneumoniae infection in vivo/in vitro

Summary: This study identified the natural flavonoid isorhamnetin as an inhibitor of PLY, which reduces the pathogenicity of S. pneumoniae. Isorhamnetin also decreases the formation of bacterial biofilms and attenuates PLY-mediated injury. In vivo experiments confirmed the efficacy of isorhamnetin in ameliorating lung tissue injury caused by S. pneumoniae.

Ameliorative effect of scopolamine-induced cognitive dysfunction by Fufangmuniziqi formula: The roles of alkaloids, saponins, and flavonoids

Summary: This study investigated the effects of Fufangmuniziqi formula (FFMN) on Alzheimer's disease (AD) and the role of alkaloids, flavonoids, and saponins in the process. The results showed that FFMN improved cognitive and memory impairment in the AD mouse model. Alkaloids enhanced the cholinergic system, while flavonoids and saponins attenuated neuroinflammation and oxidative stress. These findings suggest that the combination of alkaloid, flavonoid, and saponin fractions derived from FFMN is a promising anti-AD agent.

This study analyzed the phytochemical profile of the aqueous extract of M. longifolia and evaluated its acute and subacute toxicity in mice. The results showed that M. longifolia extract is rich in phenolic compounds and has no observed toxicity up to a certain dosage limit.

A study integrated metabolomics and network pharmacology to investigate the effects of Shicao in alleviating acute liver injury

A rat model of acute liver injury was established and liver function indices were assessed. Untargeted metabolomics was conducted to identify differential metabolites. Network pharmacology was used to construct a network of metabolite-reaction-enzyme-gene constituents, and key components and hub genes were screened. Shicao showed hepatoprotective effects against acute liver injury in rats.
