Summary: JAKs are important enzymes in the JAK-STAT signaling pathway, and dysfunctional JAKs can lead to severe immune system-related diseases. Various JAK inhibitors have been developed to block abnormal JAK-STAT signaling, but drug selectivity and resistance to these inhibitors remain challenges. This review discusses the role and structure of JAKs, as well as strategies for disrupting JAK dimerization to develop novel inhibitors.

Summary: In this study, it was found that elevated levels of IFN-gamma induce ferroptosis in salivary gland epithelial cells (SGEC) in Sjogren's syndrome (SS) through JAK/STAT1 signaling, by inhibiting the function of system Xc(-). Transcriptome analysis revealed differential expression of ferroptosis-related markers in human and mouse salivary glands of SS, with upregulation of IFN-gamma and downregulation of GPX4 and AQP5. Manipulating ferroptosis or IFN-gamma signaling can alleviate or aggravate SS symptoms in mouse models.

Summary: The study found that the osteoclastogenic cytokine RANKL induced expression of GILT in osteoclast precursors in a STAT1-dependent manner, leading to high levels of GILT in mature osteoclasts, with further enhancement by gamma-interferon. GILT, along with cathepsin K, played a role in osteoclastic resorption, as demonstrated by reduced bone-resorbing capacity in GILT-deficient osteoclasts. GILT was shown to directly reduce noncollagenous bone matrix protein SPARC and enhance collagen degradation by cathepsin K, contributing to osteoclast-mediated bone resorption.
