Summary: TAK-071, a novel M1 PAM with low cooperativity, showed improvements in sociability deficits and cognitive function in experimental animals and a genetic mouse model of schizophrenia. When combined with currently prescribed antipsychotics, it demonstrated positive effects, making it a potential new therapy for schizophrenia.

Summary: The N-Aβ core can reverse impaired synaptic plasticity in hippocampal slices from 5xFAD APP/PS1 model mice, including effects on LTP and LTD, correlating with alleviation of downregulation of hippocampal AMPA-type glutamate receptors in preparations from 5xFAD mice. The action of the N-Aβ core depends on a critical di-histidine sequence and involves the phosphoinositide-3 (PI3) kinase pathway via mTOR (mammalian target of rapamycin).

Summary: Research suggests that negative symptoms may be related to decreased concentrations of various metabolites in the prefrontal cortex, and resveratrol can improve social interaction behaviors and certain metabolite levels in a schizophrenia model.

Summary: This study investigated the prevalence of alexithymia in patients with schizophrenia and found that alexithymia mediated the relationship between cognitive deficits and negative symptoms. The results suggest that targeting emotion processing and cognition may be an effective way to alleviate negative symptoms.

Summary: This study suggests that using M1 mAChR positive allosteric modulator (PAM) VU0486846 can improve cognitive function and reduce Aβ oligomer production in female mice with Alzheimer's disease (AD). This treatment approach could be a viable disease-modifying strategy to slow down the progression of AD in women.

Summary: Early life stress may lead to schizophrenia-like phenotypes and persistent hippocampal abnormalities. Nicotinamide may be a safe and effective treatment in adolescence to restore normal hippocampal function and prevent or ameliorate schizophrenia-like behavior.

Summary: Schizophrenia is a syndrome that requires drugs targeting muscarinic M1 and M4 receptors, rather than dopamine D2 receptors, as antipsychotic drugs do. It would be ideal to identify individuals who respond to drugs activating the CHRM1 and 4 receptors, as non-response to treatment could be restricted to specific sub-groups within the syndrome.

Summary: Autism spectrum disorder (ASD) is a heterogeneous neurodevelopmental disorder caused by various genetic and environmental factors, characterized by social interaction impairments. Research has shown that enhancing 5-HT activity can reverse social deficits in ASD mouse models, while MDMA and a 5-HT1b receptor agonist are effective in rescuing sociability deficits in multiple ASD mouse models.

Summary: This study reveals the crucial role of presynaptic neurexin-ligand cerebellin-4 (Cbln4) in long-term potentiation (LTP) at EC -> DG synapses. Binding of presynaptic Cbln4 to postsynaptic neogenin-1 enables the EC -> DG synapses to undergo LTP, but is not necessary for the establishment or function of these synapses.

Summary: Acetylcholinesterase inhibitors are currently the only FDA-approved treatment for cognitive impairments in Alzheimer's disease. This study found that novel direct-acting muscarinic cholinergic receptor agonists can improve wakefulness and sleep in aged mice, suggesting potential therapeutic benefits for aging, mild cognitive impairment, and Alzheimer's disease patients.

Summary: Using a mouse model of Huntington's disease, the authors found increased connectivity in the corticostriatal pathway due to deficient cholinergic transmission. Lowering mutant huntingtin specifically in striatal cholinergic interneurons normalized the connectivity, providing evidence for a key node in the network underlying corticostriatal pathophysiology in Huntington's disease.
