Summary: In this study, a mesophilic beta-glucanase from Bacillus terquilensis CGX 5-1 was engineered to improve its stability at acidic environment and catalytic property. The Q1E/I133L variant showed better acidic stability and higher catalytic activity, with potential applications in industries. The improved acidic resistance ability of the variant may be attributed to more surface negative charge, decreased exposure degree of residue No.1, shifted side chain direction of residue No.133, and lower total and folding free energy.

The enzyme belongs to GH16 family and shares homology with an enzyme from Streptomyces sioyaensis. Mutations in disulfide bonds affect thermostability and activity, with one mutant showing increased activity possibly due to enhanced dynamics near the active site. This study provides insights into balancing rigidity and flexibility for enzyme activity, aiding in protein engineering efforts.

Summary: This study presents the catalytic sequential hydrosilylation of 1,3-enynes and 1,4-enynes promoted by cobalt complexes derived from bisphosphines. The reactions resulted in the formation of enantioenriched cyclic alkenylsilanes with simultaneous construction of a carbon-stereogenic center and a silicon-stereogenic center. The functionalization of these chiral silanes provides access to a variety of difficult-to-access chiral building blocks.

Summary: Members of the genus Bifidobacterium are difficult to genetically manipulate due to their extensive and variable Restriction-Modification systems. A modified suicide vector, pFREM28, was developed to target specific genes in B. breve by removing known restriction sites, reducing the time, effort, and resources required for generating site-directed mutants. This approach could be applied to other (bifido)bacterial species as well.

Summary: In this study, a sequential inter- and intramolecular addition reaction on electron-deficient 1,3-diynes conjugated with a ketone, ester, or imide functionality is described. This process is effectively directed by a propargylic -OH or -NH functionality. Alcohols, amines, and thiols show excellent reactivity for the initial addition. The addition of amines occurs at ambient temperature, whereas other nucleophiles require high temperatures and a base additive.

Summary: The study isolated the strain Halomonas meridiana ES021, which produces extracellular beta-1,3-1,4-glucanase, from the Red Sea off coast of Gabal El-Zeit, Egypt. The extracellular enzyme was partially purified and found to have a molecular mass of approximately 72 kDa. The purified enzyme showed strong antibacterial and antifungal effects, and also exhibited effective clarification of different juices. This is the first report of beta-1,3-1,4-glucanase enzyme from the halotolerant Halomonas species.

Summary: Through site-directed mutagenesis, a robust mutant Bifidobacterium thermophilum (BtAS) strain BtAS(Y414F/P200R) with enhanced turanose productivity was successfully developed, showing an increase in turanose yield by two times compared to the wild type BtAS and completing the reaction in half the time.

Summary: This study characterized a new alpha-amylase from Bacillus cereus GL96 and used an in-silico approach to redesign the enzyme for improved thermal stability. The engineered enzyme showed higher stability and activity at elevated temperatures compared to the wild-type enzyme. Specifically, the D162K mutation resulted in the best performance, with increased optimum temperature and improved stability. This mutant enzyme holds promise for further investigation and industrial applications.

Summary: This study explored the key enzymes involved in cucurbitacins biosynthesis in the Chinese traditional medicinal plant Hemsleya chinensis. Using AlphaFold2 and molecular docking, the researchers successfully predicted the 3D structure of HcOSC6 and its binding features to 2,3-oxidosqualene. Mutagenesis experiments identified three key amino acid residues, E246, M261, and D490, which significantly affected the activity of HcOSC6. This study not only characterized three potential key residues for producing cucurbitacins, but also provided insights into their significant role in metabolic engineering.

Summary: The study focused on the impact of the enzyme endo-beta-1,3-1,4-glucanase from Bacillus subtilis 168 on improving the nutritional and bioactive properties of barley beta-glucan. A new procedure for isolating pure beta-glucan from barley flour was developed, enabling it to be free from starch contamination. The cloned enzyme showed high efficiency in degrading beta-glucan and increasing its antioxidant capacities, suggesting potential applications in functional foods.
