Summary: The expression of Sox17 gene is crucial for the differentiation of both endothelial and endodermal cells. The study reveals that two evolutionarily conserved promoter regions (CRs) have different roles in regulating the Sox17 gene. Deletion of CR1 only has a modest effect on lympho-vasculogenesis, while deletion of CR2 impairs both vascular and endodermal development.

Summary: Drug-resistance is a major problem in multiple myeloma (MM) patients. This study reveals that activated-leukocyte-cell-adhesion-molecule (ALCAM) regulates MM side population (SP)-mediated drug-resistance through the ALCAM-EGF/EGFR axis. EGFR activation promotes the SP ratio, while ALCAM inhibits EGFR downstream signaling in MM cells. SP MM cells have a higher number of mitochondria and interference of mitochondrial function inhibits SP-genesis. Combination therapy with an anti-MM agent and EGFR inhibitor gefitinib improves MM therapeutic efficacy and prolongs survival in MM-bearing mice.

Summary: Through the use of hESCs model, this study found that TET proteins and NANOG play different roles in human hPGC specification compared to mice. TET proteins stimulate the expression of SOX17 through the NODAL signaling pathway and directly regulate NANOG expression at the onset of hPGCLC induction. DNMT3B-mediated hypermethylation in TKO hESCs inhibits the activation of relevant genes and SOX17/NANOG, leading to the repression of hPGCLC induction. Knockout of DNMT3B partially restores the NODAL signaling pathway and the expression of SOX17/NANOG, rescuing hPGCLC induction.

Summary: Recently, there has been increased attention on the purple tea cultivars with unique purple-colored leaves. These cultivars are enriched with anthocyanin, which provides a new perspective for studying the metabolic flux of flavonoids in tea plants. This review aims to summarize the latest progress on the composition and accumulation of anthocyanins in tea plants, with discussions on the regulation mechanism and a proposed hypothetical model for leaf color transformation during growth. Novel insights presented in this review will facilitate future in-depth studies of purple tea, providing a theoretical basis for targeted breeding programs in leaf color.

Summary: The use of DNA-binding dyes in stem cell research is significant for isolating specific cell populations. In this study, a distinct side population of cells was identified in the mouse brain using a DNA-binding dye, and these cells showed sensitivity to ABC transporter inhibitors. Furthermore, the majority of these cells expressed CD31 and were not adult neural stem cells.

Summary: Cancer stem cells (CSCs) play a crucial role in tumor growth, metastasis, and recurrence. Techniques such as staining tumor cells with Hoechst 33342 and analyzing the proportion of side population (SP) cells using flow cytometry provide convenient, fast, and cost-effective methods for the identification and purification of CSCs. Data generated from these assays can contribute to a better understanding of the stemness properties of tumor cells in response to various signals.

This highlights the importance of understanding the role of misfolded proteins in regulating CX3CR1.
