Summary: Fortilin prevents the activation of the TGF-beta 1 signaling pathway by binding to TGF-beta 1 and inhibiting its interaction with the TGF-beta RII receptor. This inhibits the phosphorylation of Smad3 and the differentiation of C3H10T1/2 mesenchymal progenitor cells to smooth muscle cells. These findings suggest that extracellular fortilin acts as a negative regulator of the TGF-beta 1 signaling pathway.

This review discusses the role of TGF-beta signaling in fibrotic organs, as well as potential therapeutic options for targeting tissue fibrosis through modulating TGF-beta signaling.

CCN2 increases T beta RII expression in VSMCs and maintains TGF-β pathway activation through an EGFR-SMAD dependent manner.

The results showed that there was no difference in circulating TGF-beta 1 levels before the time of active disease, but women with preeclampsia had higher levels during the active disease compared to normotensive controls. This was true for both early-onset/severe and late-onset/mild types of preeclampsia.
