Here we show that the C-terminal domain of LuxR activates the transcription of Aliivibrio fischeri luxICDABEG in Escherichia coli SKB178 gro(+) and E. coli OFB1111 groEL673 strains to the same level. Using affinity chromatography, we showed that GroEL binds to the N-terminal domain of LuxR, pointing to a GroEL/GroES requirement for the folding of the N-terminal domain of LuxR.

Summary: A new lux-biosensor based on Bacillus subtilis was developed for detecting DNA-tropic and oxidative stress-causing agents. The biosensors showed increased sensitivity to certain toxicants compared to E. coli-based lux-biosensors. They can be used for studying mechanisms of toxicity against different types of bacteria.

Summary: LitR acts as a master-regulator of gene expression in QS systems of bacteria from Vibrio and Aliivibrio genera, upregulating key genes such as luxI, luxCDABE, and luciferase. This upregulation is independent of luxR gene and depends on lux-box sequence in the luxI promoter.

The Mode of Action of Cyclic Monoterpenes (-)-Limonene and (+)-α-Pinene on Bacterial Cells

Summary: This study investigated the mechanisms of action of two volatile organic compounds on bacteria and found that (-)-limonene induces oxidative stress leading to DNA, protein, and membrane damage, while (+)-alpha-pinene only induces heat shock in bacteria. Additionally, it was shown that (-)-limonene completely inhibits the refolding process of heat-inactivated bacterial luciferase, while (+)-alpha-pinene partially inhibits this process in a mutant strain.

Influence of the luxR Regulatory Gene Dosage and Expression Level on the Sensitivity of the Whole-Cell Biosensor to Acyl-Homoserine Lactone

Summary: The study focused on the impact of luxR gene expression level on cell functionality in the LuxI/R QS system, showing that increasing the expression level and dosage of the regulatory gene can enhance sensitivity of the biosensor to autoinducers and reduce response time. The best parameters were achieved with an increased dosage of A. fischeri luxR, reaching a sensitivity of 30-100 pM to the autoinducer.

Biosensors to Assess the Activity of Promoters and Chaperones in Bacillus subtilis Cells

Summary: Plasmids were constructed to study the activities of promoters in Bacillus subtilis cells, showing that inducible promoters had higher responses to D(+)-xylose but were sensitive to glucose repression. Lux sensors with bacterial luciferases as reporter proteins assessed the influence of DnaKJE and TF chaperones on enzyme synthesis levels.

Summary: This study proposes a straightforward and robust approach for the markerless integration of DNA fragments encoding whole metabolic pathways into the genome of bacteria. By utilizing the Ocr antirestriction function of T7 phage, along with highly restrictive counterselection and lambda Red recombineering, the efficiency of integrating operon-sized cassettes into arbitrary genomic loci of four enterobacterial species is significantly improved, reaching 50-100%.

This review compares these mechanisms and discusses the potential to control self-assembly by switching between intermediate states.

MGL S3 Chimeric Enzyme Drives Apoptotic Death of EGFR-Dependent Cancer Cells through ERK Downregulation

Summary: Methionine dependence is a hallmark of malignant cells, and deprivation of methionine can induce cancer cell death. The genetically engineered protein MGL S3 can effectively reduce the number of cancer cells from multiple origins and induce cell death through apoptosis and downregulation of ERK activity. High expression of certain genes in the methionine metabolism pathway is associated with poor prognosis in breast cancer and neuroblastoma patients, and there is a strong correlation between EGFR dependency and DNMT3A/CBS expression in these cancers. Furthermore, MGL S3 enhances the sensitivity of H1299 cells to EGFR inhibitor gefitinib.

Genotoxic Effect of Dicyclopropanated 5-Vinyl-2-Norbornene

Summary: In this study, the genotoxic properties of dicyclopropanated 5-vinyl-2-norbornene (dcpVNB) were investigated using biosensors based on Escherichia coli and Bacillus subtilis. It was found that dcpVNB induces DNA damage, triggers SOS response and Dps expression, and activates the OxyR-mediated response to oxidative stress. The sensitivity of the biosensors based on B. subtilis and E. coli to dcpVNB was comparable, indicating similar permeability of the cell walls of Gram-positive and Gram-negative bacteria to this compound. Furthermore, structural changes in hydrocarbons can significantly alter their mode of toxicity, as demonstrated by the comparison with 2-ethyl(bicyclo[2.2.1]heptane) (EBH) lacking cyclopropanated groups.

Photorhabdus lux-operon heat shock-like regulation
