Composition of Phenolic Compounds and Glycoalkaloids α-Solanine and α-Chaconine during Commercial Potato Processing

The influence of a commercial production process for dehydrated potato flakes on the content of free phenolic compounds, total phenolics, and glycoalkaloids in potatoes during the subsequent processing steps was determined. Processing byproducts, such as potato peel (steam peeling), mashed potato residues, and side streams (blanching and cooking waters), have also been investigated. A high-performance liquid chromatography (HPLC) method was developed to separate and quantify caffeic acid, gallic acid, ferulic acid, p-coumaric acid, p-hydoxybenzoic acid, protocatechuic acid, vanillic acid, catechin, and three isomers of caffeoylquinic acid: chlorogenic, neochlorogenic and cryptochlorogenic acid. Determination of the glycoalkaloids a-solanine and alpha-chaconine was performed by using a high-performance thin-layer chromatography (HPTLC) method. The deliverables reveal that processing potatoes to potato flakes remarkably diminishes the content of the analyzed compounds, mainly due to peeling and leaching. The influence of thermal exposure is less significant. About 43% of the initial phenolic acids and 10% of the glycoalkaloids remain after processing. The results of the total phenolic content assay by Folin-Ciocalteu reagent are proportional to the content of phenolic compounds determined by HPLC. Steam peeling has a higher influence on glycoalkaloid losses compared to that on phenolics. The highest amounts of phenolic compounds and glycoalkaloids were found in peeling byproduct. During processing, the amount of chlorogenic acid decreased, whereas the concentration of neochlorogenic acid increased due to isomerization. The impact of the results on potato processing technology is discussed.

Summary: The study developed a proteomics strategy to establish specific markers for quantifying proteins in honey, especially for distinguishing honeydew honeys from floral honeys. By quantifying selected marker peptides, such as QNIDVVAR, the study showed the potential of using specific protein content as a marker for authenticating honeydew honeys. Ultimately, principal components analysis highlighted the content of QNIDVVAR as a good descriptor for distinguishing different honey samples.

Epigenetic DNA Methylation of EBI3 Modulates Human Interleukin-35 Formation via NFkB Signaling: A Promising Therapeutic Option in Ulcerative Colitis

Summary: Research has shown that upregulation of EBI3 through histone acetylation can alleviate symptoms in a mouse model of chronic colitis. Further investigation revealed that under inflammatory conditions, treatment with DNA methylation inhibitors and TNF alpha led to synergistic upregulation of EBI3 in human colon epithelial cells. The results suggest that the upregulation of EBI3 under inflammatory and demethylating conditions may lead to the formation of anti-inflammatory IL-35, which could be a potential therapeutic target for colitis.

Different extraction and digestion protocols were optimized for different nuts, with SDS buffer extraction suitable for almond, peanut, and walnut samples, and PVPP buffer extraction recommended for cashew, pistachio, and hazelnut samples. Various methods were assessed for protein extraction and digestion efficiency in the study.

Activated carbons prepared from spent coffee grounds and parchment showed lower adsorption capacity for lactic acid compared to standard commercial granular activated carbon. These results demonstrate the potential of using coffee by-products as low-cost adsorbents for removing organic compounds from aqueous solutions.

Summary: In this study, a simplified approach based on reverse phase and ion exchange chromatography was utilized to purify the major wheat alpha-amylase/trypsin inhibitors (ATIs). The efficiency of the purification process was demonstrated by obtaining pure fractions of the targeted ATIs with high purity levels. These purified ATIs can now be further studied to gain more knowledge about their involvement in non-celiac sensitivity.

Targeted Bottom-Up Mass Spectrometry Approach for the Relative Quantification of Post-Translational Modification of Bovine κ-Casein during Milk Fermentation

Summary: This study investigated the post-translational modification (PTM) of kappa-casein (kappa-CN) during bovine milk fermentation. A LC-MS method was developed to identify and quantify the modified and unmodified variants of four peptides containing serine and threonine residues. The results showed that over 50% of kappa-CN existed in one of its modified forms in milk, and fermentation significantly altered the composition of unmodified/modified kappa-CN.

Limited enzymatic hydrolysis of green coffee protein as a technique for preparing new functional food components

Summary: The objective of this study was to improve the functional properties of green coffee protein. Extraction of defatted green coffee meal using PVPP increased protein yield and decreased the antioxidant activity of the meal. Peptic hydrolysis significantly increased the degree of hydrolysis, and enzymatic hydrolysis improved the antioxidant activity of the green coffee protein. The results suggest that enzymatic hydrolysis with pepsin is an effective technique to obtain bioactive compounds.

Development of Non-Targeted Mass Spectrometry Method for Distinguishing Spelt and Wheat

Summary: Food fraud is widespread and requires innovative strategies to combat it. This study introduces a new non-targeted method (NTM) to distinguish between spelt and wheat, which is helpful in detecting fraud and testing authenticity. By using liquid chromatography coupled high-resolution mass spectrometry (LC-HRMS), a highly resolved fingerprint in the form of spectra is obtained for various cultivars of spelt and wheat. Convolutional neural network (CNN) models are built and trained using duplicate measurements of eleven cultivars, allowing them to automatically learn patterns and accurately classify samples into spelt or wheat. The results demonstrate the reliability of this approach on a wider range of cultivars.
