Summary: We investigated the relationship between the cognitive status of participants' spouses and participants' own cognitive outcomes, controlling for mid-life factors. Results showed that having a spouse with MCI/dementia was associated with a deficit in cognitive function, but this association was attenuated after adjusting for mid-life risk factors. There was no significant relationship between spousal MCI/dementia status and incident dementia.

Summary: The study developed a population pharmacokinetic (PK) and pharmacodynamic (PD) model for crizanlizumab, a monoclonal antibody for sickle cell disease. The results showed that crizanlizumab exhibited nonlinear PK and factors such as renal and hepatic impairments, concomitant hydroxyurea usage, and anti-drug antibody did not impact its exposure. The model can be used to extrapolate PK to pediatric population and evaluate alternative regimens and routes of administration.

Additionally, combining VCAM-1 and cerebrospinal fluid (CSF) biomarkers showed independent associations with AD-related clinical impairment.

Summary: When evaluating EpCAM-based enrichment technologies for CTCs, it is important to use cell lines that closely resemble real CTCs and measure the EpCAM expression of both CTCs and cell lines at different institutions and times. In this study, CTCs were enriched by depleting leukocytes from diagnostic leukapheresis products and EpCAM expression was measured using quantitative flow cytometry. Results showed varying but relatively low EpCAM expression in CTCs from castration-sensitive prostate cancer patients. There were also significant variations in antigen expression among identical cell lines cultured at different institutions, affecting capture efficiency.

Summary: This study investigated the effect of bortezomib (BTZ) on vascular permeability and expression of cell-cell junction molecules in human pulmonary microvascular endothelial cells. The findings showed that clinically relevant concentrations of BTZ increased the permeability of endothelial cell monolayers and downregulated the expression of claudin-5, occludin, and VE-cadherin at both protein and mRNA levels. This suggests that BTZ-induced pulmonary toxicity may be mediated by increased vascular permeability caused by the downregulation of these cell-cell junction molecules.

Summary: Vascular cell adhesion molecule-1 (VCAM-1) plays a crucial role in atherosclerosis and has been targeted for anti-atherosclerotic drug development. Most current VCAM-1-inhibiting drug candidates indirectly down-regulate VCAM-1 expression by inhibiting upstream cytokines, but optimizing dosage to balance pathological activity and normal physiological function is challenging. Recently, specific strategies that selectively inhibit VCAM-1 function, such as peptide- and antibody-based therapeutics, have emerged. However, studies on these novel VCAM-1-directed therapies are limited by the complexity of atherosclerosis pathophysiology.

The study found that adult-child caregivers reported higher burden levels, even after controlling for demographic factors and location of care. Social workers should consider tailored strategies to address and reduce burden for adult-child caregivers, beyond typical interventions for spousal caregivers.

Summary: DSCAM, a member of the immunoglobulin superfamily, plays crucial roles in neural development by mediating cell adhesion and self-avoidance of neurites. Through electron microscopy and additional methods, it has been found that DSCAM forms a regular pattern at cell adhesion interfaces, with Ig-like domains contributing to interactions and FnIII domains crucial for pattern formation and cell membrane interaction. On the other hand, no obvious assembly pattern is observed with other DSCAMs, indicating different structural roles in cell adhesion and neural network formation.

Summary: This study investigates the role of steroid treatment in identifying cancer patients who are distressed or mentally disordered. By analyzing the medical records of 12,298 cancer patients, the study identifies four subgroups of patients using different dosages of prednisone equivalents. The study finds that patients with higher dosages of prednisone are more likely to receive psychotropic drug administration, while patients with lower dosages have a slightly increased probability of psychiatric assessment and psychotropic drug administration. The subgroup with the lowest probability of receiving steroid treatment also has the lowest likelihood of receiving psychiatric assessment and psychotropic drug administration.

Summary: International guidelines recommend screening for cancer patients’ distress, but rates are insufficient. This study developed a survey to assess systemic barriers to distress screening. Through a three-step approach, an initial survey with 53 questions was reduced to a final set of 14 validated questions using qualitative content analysis and factor analysis. Major barriers identified include timing of screening, lack of capacity, patient and staff overload, and refusal of patients to be referred to support services. The validated questions enable quick identification and improvement of screening programs.

Summary: The COVID-19 pandemic has negatively affected the mental health of ICD patients, leading to fear, anxiety, depression, and PTSD. Factors such as unemployment, younger age, low social support, prior COVID-19 infection, and inadequate knowledge about COVID-19 predicted these mental health issues. Fear of COVID-19 had an impact on anxiety, depression, and PTSD, with positive and negative affects mediating these relationships. Psychological and educational interventions are crucial for vulnerable subgroups of ICD patients.

The retina is an extension of the brain and retinal symptoms in subjects with neurodegenerative diseases substantiated the eye as a window to the brain. The developed model has a high sensitivity in detecting patients who decline psycho-oncological support, with older patients, patients with lower distress levels, fewer comorbidities, fewer physical problems, and those who do not feel sad, afraid, or worried being more likely to refuse psycho-oncological support.

Summary: This study comprehensively characterized a pair of monozygotic female twins, one of whom experienced TTS. The twin sister with TTS exhibited higher levels of anxiety, vital exhaustion, social inhibition, and alexithymia, and lower levels of quality of and meaning in life. Additionally, the twin with TTS experienced more-and also more severe-stressors involving life-threatening and dangerous situations over the life course.

Changes in FA from pre- to post-treatment were associated with relevant outcome variables at post-treatment and 12-month follow-up.
