Summary: Liver cancer chemotherapy is limited by high toxicity and poor specificity, but targeted drug delivery systems have emerged as a novel therapeutic strategy for delivering precise drug doses to target sites. In this study, DTX/GA-HA-NPs were successfully prepared with high monodispersity and desirable particle size, showing strong inhibitory effects on HepG2 cell proliferation and promoting apoptosis. The results suggest promising liver-targeting and antitumor activities of DTX/GA-HA-NPs in vitro, laying the foundation for future in vivo studies.

Therefore, researchers have designed HA-based drug delivery platforms for CD44-mediated tumor targeting. HA-based nanoprodrugs possess distinct advantages such as good bioavailability, long circulation time, and controlled drug release and retention ability, and have been extensively studied in the past years.

Summary: Dual-receptor targeted (DRT) nanoparticles with two distinct targeting agents were prepared for the delivery of docetaxel (DTX) to EGFR and PD-L1 positive cancer cells. The DRT-DTX-PLGA nanoparticles exhibited high cytotoxicity and enhanced apoptotic cell death compared to single ligand-targeted nanoparticles. The dual receptor mediated endocytosis of DRT-DTX-PLGA resulted in high intracellular DTX concentration and cytotoxic properties. DRT nanoparticles have the potential to improve cancer therapy by providing selectivity over single-ligand-targeted nanoparticles.

Summary: Radiation therapy is commonly used in the treatment of tumors, but it often causes damage to normal tissues. Boron neutron capture therapy (BNCT) is a targeted approach that has shown advantages in tumor treatment. This study developed a tumor-targeting nano-boron drug by using albumin as a drug carrier and modifying the nanoparticles with hyaluronic acid. The results showed that the modified nanoparticles effectively delayed drug release, enhanced aggregation in tumors, and had good safety with no noticeable toxicity. These findings provide valuable insights for the application of boron drugs in BNCT.

Summary: Colon-targeted trans-cinnamic acid (tCA) and its conjugates with glutamic acid (tCA-GA) and aspartic acid (tCA-AA) were designed and synthesized. The anti-colitic activity of colon-targeted tCA was evaluated using a rat colitis model. The conjugates decreased the distribution coefficient and cell permeability of tCA and produced more tCA in the cecum, particularly with tCA-GA. Oral administration of tCA-GA delivered a higher amount of tCA to the cecum and exhibited better anti-colitic effects compared to tCA and the current treatment, sulfasalazine (SSZ).

Summary: The study presents the design of a tailored structure for renal-clearable zwitterionic cyclodextrin for selective drug delivery in colorectal cancer. Twenty cyclodextrin derivatives were synthesized and screened for colloidal stability, resulting in five candidates that were evaluated for biodistribution and an optimized structure was identified. The optimized cyclodextrin demonstrated high tumour accumulation and improved antitumour efficacy in colorectal cancer models.

SAR demonstrated better colon specificity, anticolitic potency, and reduced systemic absorption compared to RAS, making it a potentially safer alternative.

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
