Development of poly(lactic-co-glycolic) acid nanoparticles-embedded hyaluronic acid-ceramide-based nanostructure for tumor-targeted drug delivery

A hyaluronic acid-ceramide (HACE) nanostructure embedded with docetaxel (DCT)-loaded poly(D,L-lactide-co-glycolide) (PLGA) nanoparticles (NPs) was fabricated for tumor-targeted drug delivery. NPs with a narrow size distribution and negative zeta potential were prepared by embedding DCT-loaded PLGA NPs into a HACE nanostructure (DCT/PLGA/HACE). DCT-loaded PLGA and DCT/PLGA/HACE NPs were characterized by solid-state techniques, including Fourier-transform infrared (FT-IR) spectroscopy, differential scanning calorimetry (DSC), and powder X-ray diffraction (PXRD). A sustained drug release pattern from the NPs developed was observed and negligible cytotoxicity was seen in NIH3T3 cells (normal fibroblast, CD44 receptor negative) and MDA-MB-231 cells (breast cancer cells, CD44 receptor positive). PLGA/HACE NPs containing coumarin 6, used as a fluorescent dye, exhibited improved cellular uptake efficiency, based on the HA-CD44 receptor interaction, compared to plain PLGA NPs. Cyanine 5.5 (Cy5.5)-labeled PLGA/HACE NPs were injected intravenously into a MDA-MB-231 tumor xenograft mouse model and demonstrated enhanced tumor targetability, compared with Cy5.5-PLGA NPs, according to a near-infrared fluorescence (NIRF) imaging study. Summary: An HAase inhibitor-incorporated hyaluronic acid hydrogel was designed to reduce toxicity risk and slow biodegradation rate in subcutaneous tissue. The hydrogel, formulated with Quercetin as an HAase inhibitor and quetiapine as a model drug, exhibited sustained drug release and improved pharmacokinetic parameters in rats following subcutaneous injection. All results supported the designed hydrogel as a biocompatible formulation for sustained drug delivery.

Determination of Leuprolide-Fatty Acid Conjugate in Rat Plasma Using LC-MS/MS and Its Pharmacokinetics after Subcutaneous Administration in Rats

Summary: In this study, a simple UPLC-MS/MS method was developed for the simultaneous determination of leuprolide and leuprolide-oleic acid conjugate (LOC) levels. The pharmacokinetics of leuprolide and LOCs were evaluated and it was found that conjugation with fatty acids significantly altered the pharmacokinetics of leuprolide.

Factors determining the oral absorption and systemic disposition of zeaxanthin in rats: in vitro, in situ, and in vivo evaluations

Summary: This study investigates the oral absorption and systemic disposition of zeaxanthin. The results show that zeaxanthin has poor solubility and stability in simulated gut luminal fluids. It is primarily absorbed in the duodenum, but a large fraction remains unabsorbed. Zeaxanthin accumulates in the liver, lung, and spleen after intravenous injection, with the liver being the major eliminating organ.

Colon-Targeted Trans-Cinnamic Acid Ameliorates Rat Colitis by Activating GPR109A

Summary: Colon-targeted trans-cinnamic acid (tCA) and its conjugates with glutamic acid (tCA-GA) and aspartic acid (tCA-AA) were designed and synthesized. The anti-colitic activity of colon-targeted tCA was evaluated using a rat colitis model. The conjugates decreased the distribution coefficient and cell permeability of tCA and produced more tCA in the cecum, particularly with tCA-GA. Oral administration of tCA-GA delivered a higher amount of tCA to the cecum and exhibited better anti-colitic effects compared to tCA and the current treatment, sulfasalazine (SSZ).

Tailoring renal-clearable zwitterionic cyclodextrin for colorectal cancer-selective drug delivery

Summary: The study presents the design of a tailored structure for renal-clearable zwitterionic cyclodextrin for selective drug delivery in colorectal cancer. Twenty cyclodextrin derivatives were synthesized and screened for colloidal stability, resulting in five candidates that were evaluated for biodistribution and an optimized structure was identified. The optimized cyclodextrin demonstrated high tumour accumulation and improved antitumour efficacy in colorectal cancer models.

N-Succinylaspartic-Acid-Conjugated Riluzole Is a Safe and Potent Colon-Targeted Prodrug of Riluzole against DNBS-Induced Rat Colitis

SAR demonstrated better colon specificity, anticolitic potency, and reduced systemic absorption compared to RAS, making it a potentially safer alternative.

3D human foreskin model for testing topical formulations of sildenafil citrate

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
