Summary: The main objective of this research was to prepare a nanosuspension (ChlE-NS) of the chloroform extract (ChlE) of the Egyptian marine red macroalga D. simplex to improve its aqueous solubility and bioactivity. The results showed that ChlE-NS exhibited greater cytotoxic activity against cancer cells compared to ChlE, and also had improved antioxidant activity.

Summary: A library of novel 1,3,4-oxadiazole bioisosteres was synthesized and evaluated for their cytotoxic activity. Several of the new compounds showed potent anticancer activity, surpassing the previously synthesized oxadiazole compound. The nature of the selenadiazole and thiadiazole rings may play a crucial role in the antitumor activity. These compounds exhibited strong cytotoxic effects in tumor cells compared to human primary cells.

Summary: Resveratrol, a natural compound with antioxidant properties, has potential as both a chemopreventive and cancer chemotherapeutic agent. Due to its poor bioavailability, the study of new derivatives is important. In this study, (E)-stilbenes derived from resveratrol and cyclic analogues with benzofuran or indole nucleus were synthesized and evaluated for their impact on tumor growth. Some compounds showed higher cytotoxicity than (E)-resveratrol, with one derivative demonstrating eight times greater cytotoxic activity against colon cancer cells. Certain cyclic derivatives also exhibited strong inhibition of cell growth.

Summary: This work demonstrates the use of a titration method to produce multilayered coatings on a nanosuspension, enhancing the release behavior and stability of curcumin. The results show that a pH-responsive outer layer delays curcumin release in acidic conditions and facilitates its release in neutral conditions. Increasing the number of layers improves the stability of curcumin in the nanosuspension and reduces its toxicity.

Summary: A new lanostane-type triterpene, 3-oxo-5 alpha-lanosta-7,9(11)-dien-24-oic acid methyl ester (2), and three known compounds were isolated from the fruiting bodies of Ganoderma lucidum. The cytotoxic activity of these compounds against cancer cell lines was evaluated, and it was found that compound 1 exhibited strong cytotoxicity against SK-Hep-1, HepG2, and Hela cells, while compound 4 showed moderate cytotoxicity against Hela and Hela/VCR cells.

Summary: In vitro and in vivo studies demonstrated the antitumor effects of TTE on colon tumor cell lines and a murine colorectal cancer model, including suppression of cell growth, migration, and angiogenesis, as well as promotion of antitumor immunogenic cell death. Transcription profiling and pathway analysis revealed that TTE triggers specific gene expression and autophagy stress pathways, leading to its antitumor effects.

Summary: This study developed two different nanoformulations, nanosuspension and self-nanoemulsifying drug delivery systems, and evaluated their potential in treating type 2 diabetes. The results showed that both formulations significantly reduced lipid/cholesterol and blood glucose levels, and increased antioxidant levels. The combination of the drugs in the nanoformulations was more effective, possibly due to the dual benefits of reducing insulin resistance and controlling blood glucose levels. The nanosuspension showed better oral bioavailability enhancement compared to the self-nanoemulsifying drug delivery systems.

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
