Effect of Oven and Microwave Heating on the Total Antioxidant Capacity of Dietary Onions Grown in Turkey

The active components of largely consumed dietary onions (Allium species) in Turkey were extracted with water and ethanol, separately heated in a drying oven (at 50 degrees C, for 14 h) and in a microwave oven (at 90 W, for 14 min), and their total antioxidant capacity was determined with different electron transfer-based assays. Five different onion species/aerial parts, namely yellow, red, white, fresh green leaf, and fresh green root, were measured for total antioxidant capacity with different methods, the hierarchic order in aqueous extracts being: CERAC: yellow-skinned > red-skinned > spring-root > spring-leaf > white-skinned onion; CUPRAC: spring-leaf > red-skinned > spring-root > yellow-skinned > white-skinned onion; Folin-Ciocalteau method: spring-leaf > spring-root > red-skinned > yellow-skinned > white-skinned onion. Using all three methods, white onion, showed the lowest total antioxidant capacity, while with respect to two methods (i.e., CUPRAC and Folin-Ciocalteau), spring onion-leaf showed the highest value. In ethanolic extracts, white onion exhibited the lowest total antioxidant capacity using two methods (CUPRAC and Folin-Ciocalteau). Of the heat-processed onions, the highest CERAC and Folin-Ciocalteau total antioxidant capacity values were obtained for red-skinned onions, while the highest CUPRAC value was for spring onion leaves. All three assays marked white-skinned onion as the lowest total antioxidant capacity content of heat-processed products. The change in total antioxidant capacity caused by both heating processes was not drastic; spring onion leaves essentially maintained its total antioxidant capacity level after 4 min microwave or 4 h drying oven heating. Onion processing by heat treatment did not cause a drastic loss in antioxidant values, favourable for traditional cooking practices.

Gold-Nanoparticle Based Turn-on Fluorometric Sensor for Quantification of Sulfhydryl and Disulfide Forms of Biothiols: Measurement of Thiol/Disulfide Homeostasis

Summary: This study developed a method for determining biothiols in sulfhydryl and disulfide forms using a fluorometric sensor composed of citrate-stabilized gold nanoparticles and Rhodamine-6G dye. The method can differentiate between oxidized and reduced thiols, measure thiol/disulfide homeostasis, and is selective over potential interfering compounds. It also allows for naked-eye detection of biothiols and can be applied to pharmaceutical samples containing thiols.

Peroxyl radical scavenging activity measurement of antioxidants using histidine-stabilized and glutathione-capped fluorescent gold nanoclusters

Summary: A fluorescent gold nanocluster was used to determine the peroxyl radical scavenging activity of antioxidants. The addition of glutathione enhanced the fluorescence intensity of the gold nanoclusters, while antioxidants reduced the decline in fluorescence intensity caused by peroxyl radical attack. This method provides a simple and selective estimation of peroxyl radical scavenging activity in complex matrices.

Voltammetric Measurement of Antioxidant Activity by Prevention of Cu(II)-Induced Oxidative Damage on DNA Bases Using a Modified Electrode

Summary: In this study, a gold nanoparticle (AuNP)-decorated and multiwalled carbon nanotube (MWCNT)-Nafion-modified glassy carbon electrode (GCE/AuNP/MWCNT-Nafion) was developed to assess the preventive effect of antioxidants on oxidative DNA damage. The electrochemical method relied on two phenomena: (i) reactive species (RS) produced by dissolved oxygen in the presence of copper(II) partially damaged the DNA immobilized on the electrode surface, and (ii) antioxidant compounds prevented this damage by scavenging the formed RS. The DNA protective ability of various antioxidant compounds was evaluated, with quercetin showing the highest antioxidant effect.

Spectrophotometric Fluoride Determination Using St. John's Wort Extract as a Green Chromogenic Complexant for Al(III)

Summary: In this study, a novel and eco-friendly chromogenic agent for fluoride determination was developed using green analytical chemistry. The method made use of a non-toxic Al(III)-flavonoid complex from St. John's wort plant extract. The method showed a linear response within the F- concentration range of 0.11-1.32 mM, with low LOD and LOQ values that met the permissible limits for fluoride in water. Interference effects from common anions and cations were investigated and easily eliminated with specific techniques. The method was successfully applied to real water samples and validated against a reference spectroscopic method.

Heteroatom-Doped Carbon Quantum Dots and Polymer Composite as Dual-Mode Nanoprobe for Fluorometric and Colorimetric Determination of Picric Acid

Summary: Oxygen- and nitrogen-heteroatom-doped carbon dots were prepared for the selective and sensitive determination of picric acid. The fluorescence of the carbon dots was selectively quenched by picric acid through ground-state interaction. The detection limits for picric acid were 12.5 pM (by emission measurement) and 0.9 nM (by absorption measurement). Smartphone-assisted fluorometric detection of picric acid was also achieved by incorporating the carbon dots into a polymer composite film.

A novel electrochemical sensor based on phosphate-stabilized poly-caffeic acid film in combination with graphene nanosheets for sensitive determination of nitro-aromatic energetic materials

Summary: This work presents a novel sensor electrode for electrocatalytic reduction of nitro-aromatic explosives (NAEs) by combining electrochemically reduced graphene nanosheets (GNSs) with a poly-caffeic acid (pCAF) film-modified glassy carbon electrode (GCE). The sensor showed high performance for TNT detection and could accurately determine various NAEs. The optimized electrode demonstrated resistance to interference effects and satisfactory results in real samples.
