Real-time polymerase chain reaction for the quantitative detection of tetA and tetB bacterial tetracycline resistance genes in food

A new, rapid, sensitive and specific method was developed to directly detect and quantify tetA and tetB in food. Both tet genes are two of the most frequently present tetracycline resistance genes in Gram-negative bacteria. A set of primers and Taqman probes was designed for each gene. The standard curves were performed using Escherichia coli BM13 (C600 RifR)/RP4 and E. coli NCTC 50365, which carry tetA and tetB, respectively. Meat and fish samples inoculated with these reference strains were used as a matrix to construct the standard curves for the analysis of 20 samples of chicken meat and 10 samples of hake (Merlucius merlucius). The limits of detection in pure culture were 5 cfu/mL (0.7 log cfu/mL) in the case of tetA. 50 cfu/mL (1.7 log cfu/mL) for tetB and 5 x 10(2) cfu/g (2.7 log cfu/g) for both genes in food samples. The results obtained by real-time quantitative polymerase chain reaction (qPCR) were compared to counts of tetracycline-resistant bacteria obtained by plating extracts of poultry and hake samples in culture media supplemented with 16 mg/L of tetracycline. Counts of tetracycline-resistant bacteria obtained by qPCR showed a positive correlation, especially interesting when compared with microbiological counts of tetracycline-resistant Enterobacteriaceae in poultry meat (r = 0.5509) and with tetracycline-resistant mesophilic aerobic bacteria in hake samples (r = 0.7146). The obtained results demonstrate that this method could be a useful tool for the direct quantification of the amount of bacterial strains that carry tetA and/or tetB genes in food samples. Identification and Quantification of 29 Active Substances by HPLC-ESI-MS/MS in Lyophilized Swine Manure Samples

Summary: Veterinary drugs are commonly used in food-producing animals to prevent and treat diseases. Analyzing pharmaceutical residues in animal manure can help evaluate legal and illegal practices in food production without harming animals. A method for simultaneously analyzing 29 active substances in manure samples was described in this article, with antibiotics and antiparasitic agents being the main focus. The method was validated and showed potential for controlling the administration of pharmaceuticals in food-producing animals.

Retention and Determination of Polycyclic Aromatic Hydrocarbons from Urban Air Based on Recycled Polyurethane Foam Modified with Expanded Polystyrene

Summary: This study proposes to use recycled EPS to modify the surface of PUFs for the retention of 12 PAHs in air samples. The modified material is suitable for the retention-elution of PAHs from air samples and can be used in liquid chromatography for quantitative analysis. Eighteen urban air samples were collected and analyzed weekly, and a relationship between meteorological parameters and PAH concentration was observed using PCA.

Effect of Partial Meat Replacement by Hibiscus sabdariffa By-Product and Pleurotus djamor Powder on the Quality of Beef Patties

Summary: The effects of adding Hibiscus sabdariffa by-product (2-5%) and Pleurotus djamor powder (5-7.5%) as meat replacers to beef patties were analyzed. The addition of these non-meat ingredients decreased moisture and increased fiber content, without affecting the protein level. While the roselle by-product had limited antioxidant effect, Pleurotus djamor favored oxidation processes. The samples with these ingredients had lower microbial counts, but were significantly affected in terms of texture and sensorial parameters.

The Human Gut Virome and Its Relationship with Nontransmissible Chronic Diseases

Summary: The human gastrointestinal tract contains diverse microorganisms that interact with the host and play important roles in regulating metabolic processes. While the gut bacteriome has been extensively studied, there is increasing interest in understanding the influence of other microbial groups, such as the gut virome and phages, on the host immune system and metabolic functions. Investigating the role of virome-rectification-based therapies in various diseases is also important. In future research, stool metagenomic investigations should focus on identifying bacteria and phages, as well as studying their correlation networks, to gain a better understanding of gut microbiota activity in metabolic disease progression.

Kinetic Models of Disinfection with Sodium Hypochlorite and Peracetic Acid of Bacteria Isolated from the Effluent of a WWTP

Antioxidant and Antimicrobial Effect of Biodegradable Films Containing Pitaya (Stenocereus thurberi) Extracts during the Refrigerated Storage of Fish

Summary: This study investigated the use of pitaya extracts in biodegradable packing films to inhibit quality loss in fish muscle during refrigerated storage. The experiments utilized pitaya-gelatin films for hake muscle storage and pitaya-polylactic acid (PLA) films for Atlantic mackerel muscle storage. The results showed that the presence of pitaya extract led to a significant inhibitory effect on lipid damage, microbial activity development, and growth of bacteria in both systems. The preservation effects were attributed to the presence of betalains and phenolic compounds in the pitaya extracts.

Compliance with Nutritional Recommendations and Gut Microbiota Profile in Galician Overweight/Obese and Normal-Weight Individuals

Summary: Various studies have found that specific groups or dietary compounds play a role in the onset and progression of obesity, and gut microbiota serves as a mediator between these compounds and the associated inflammation. This study aimed to evaluate the dietary intake of overweight, obese, and normal-weight individuals and analyze their gut microbiota profile in relation to changes in Body Mass Index (BMI) and diet. The results showed significant differences in fiber and monounsaturated fatty acids (MUFAs) intake among different BMI groups, with better intake adequacy observed in the normal-weight group. The analysis of gut microbiota revealed distinct microbial composition in overweight and obese individuals compared to normal-weight individuals, with specific genera identified as biomarkers for each group.

The Evaluation of Cellulose Acetate Capsules Functionalized for the Removal of Cd(II)

Summary: Cellulose acetate, derived from cellulose, is biodegradable and reusable. Functionalized capsules with Cyanex 923 were able to effectively remove and recover cadmium. Different variables, such as HCl concentration and carrier content, influenced the extraction efficiency of cadmium. The study also tested three isotherm models for modeling the cadmium removal process.

Protein marshmallow can serve as such a carrier due to its dietary and natural properties. This study aims to identify the dependence of technological factors on the rheological properties of the product and the growth of lactic acid microorganisms in confectionery products enriched with lyophilised strains.

Phytochemical Constituents, Antimicrobial Properties and Bioactivity of Marine Red Seaweed (Kappaphycus alvarezii) and Seagrass (Cymodocea serrulata)

Summary: This study evaluated the phytochemical constituents, antioxidant, and antibacterial properties of Kappaphycus alvarezii and Cymodocea serrulata. Both seaweeds contained flavonoids, tannins, phenolic compounds, glycosides, steroids, carbohydrates, and ashes. Alkaloids were only found in K. alvarezii. The chloroform extracts of both seaweeds exhibited antagonistic efficacy against Bacillus subtilis, Vibrio parahaemolyticus, Vibrio alginolyticus, Vibrio harveyi, and Klebsiella pneumoniae. GC-MS analysis revealed the presence of numerous bioactive compounds in both seaweeds, which contributed to their antioxidant and antibacterial activities. Based on the results, K. alvarezii and C. serrulata could be considered as valuable sources of nutritional and medicinal ingredients.

Simple and efficient removal of azo dyes from water samples by dispersive solid-phase micro-extraction employing graphene oxide with high oxygen content

Summary: In this study, a technique involving dispersive solid-phase micro-extraction (DSP mu E) and graphene oxide (GO) particles with a higher oxygen content synthesized through the modification of the Hummers method was investigated for the removal of azo dyes from tap water samples. The optimal conditions for the removal of the dyes were determined, and the precision of the method was evaluated. The proposed method showed high efficiency in removing the azo dyes from the water samples.

Influence of sampling location and processing on the assembly and network of Polygoni Multiflori Radix surface microbiome

The wooden shelf surface and cheese rind mutually exchange microbiota during the traditional ripening process

Reduction of PDO Pecorino Siciliano cheese making duration: Microbial dynamics and quality attributes deriving from replacing whey permeate with hot water during cooking

Summary: This study aimed to reduce the production time of PDO Pecorino Siciliano cheese by comparing two cooking methods and assessing their impact on microbiological composition and lactic acid bacteria populations. The results showed that the different cooking methods did not affect the physicochemical characteristics of the cheese, and sensory evaluation confirmed that the modified production protocol did not significantly alter the product characteristics and overall acceptance. Therefore, cooking under hot water proved to be an effective way to reduce the transformation duration while maintaining the typicality of PDO Pecorino Siciliano cheese.

Effects of ohmic heating with different voltages on the quality and microbial diversity of cow milk during thermal treatment and subsequent cold storage

Summary: The study evaluates the effect of Ohmic heating (OH) at various voltage gradients and water bath on microbial inactivation, physicochemical and sensory properties, and microbial flora of pasteurized milk. Results show that OH with higher voltage effectively inactivates microorganisms, slows down lipid oxidation, and maintains sensory quality and amino acids content of milk. The microbial community in milk treated with OH remains relatively stable during storage.

Bacteriophages PECP14, PECP20, and their endolysins as effective biocontrol agents for Escherichia coli O157:H7 and other foodborne pathogens

Summary: In this study, two bacteriophages, PECP14 and PECP20, infecting Escherichia coli O157:H7 were isolated and found to be stable under different environmental conditions. These phages exhibited specificity in targeting different host receptors and demonstrated the ability to infect other foodborne pathogens. The efficacy of phage treatment was demonstrated through rapid adsorption and bacterial lysis. Furthermore, purified endolysins derived from these phages showed remarkable bacteriolytic activity not only against E. coli O157:H7 but also against other pathogens. These findings suggest that these phages and their endolysins can be used for biological control of E. coli O157:H7 in various stages of agricultural product production.

A systematic review on the occurrence of Salmonella in farmed Tenebrio molitor and Acheta domesticus or their derived products

Although the number of studies is limited and there are some limitations, this review highlights the importance of obtaining reliable data on the presence of Salmonella in insects, considering the growing market and increased farming scale.

Comparison of Listeria monocytogenes alternative detection methods for food microbiology official controls in Europe

Summary: Listeriosis is a severe foodborne disease with a high mortality rate, mainly caused by L. monocytogenes. The EU regulation sets qualitative or quantitative food safety criteria for L. monocytogenes and recommends the use of EN ISO 11290-1 as the reference detection method. Our study compares alternative certified methods for detecting L. monocytogenes, highlighting the importance of diversifying available detection methods.

Summary: This study detected Toxoplasma gondii DNA in raw bovine milk samples from expansion tanks in small properties located in different cities of the Midwest region of Sa similar to o Paulo, Brazil using PCR. The results showed that the TgNP1 and TgNP2 genes were more efficient in detecting T. gondii DNA, highlighting the importance of raw bovine milk as a potential source of human infections caused by this parasite, and emphasizing the need for proper pasteurization and further understanding of the epidemiology of this protozoan.

Salmonella in horses at slaughter and public health effects in Italy

Summary: This study assessed the role of equids as carriers of Salmonella and the occurrence of contaminated equid carcasses during the slaughter process. Salmonella was detected in a small percentage of equids' caecal contents, but not in carcass samples. Additionally, a genetic match was found between Salmonella strains from horse caecal contents and human cases, suggesting a link between horse meat consumption and salmonellosis.

Effect of multiple nonthermal plasma treatments of filamentous fungi on cellular phenotypic changes and phytopathogenicity

Summary: The effect of multiple sublethal doses of non-thermal plasma treatments on phenotypical changes and the reduction in phytopathogenicity of three species of fungi was investigated. The plasma damaged the surface of the fungal hyphae, decreased their dry biomass, and affected the activity of pectinase and xylanase. Furthermore, the plasma treatments increased the germination rate of artificially infected cucumber seeds and resulted in plants with higher biomass and longer roots and stems. However, the reduction in phytopathogenicity was not complete after the multiple plasma treatments.
