Reduced Systemic Levels of IL-10 Are Associated with the Severity of Obstructive Sleep Apnea and Insulin Resistance in Morbidly Obese Humans

Obstructive sleep apnea (OSA) has been related to elevation of inflammatory cytokines and development of insulin resistance in morbidly obese (MO) subjects. However, it is still unclear whether the systemic concentration of anti-inflammatory mediators is also affected in MO subjects directly related to the severity of OSA and level of insulin resistance. Normal weight and MO subjects were subjected to overnight polysomnography in order to establish the severity of OSA, according to the apnea-hypopnea index (AHI). Blood samples were obtained for estimation of total cholesterol and triglycerides, insulin, glucose, insulin resistance, tumor necrosis factor alpha (TNF-alpha), interleukin 12 (IL12), and interleukin 10 (IL-10). Serum levels of IL-10 were significantly lower in MO subjects with OSA than in MO and control individuals without OSA. Besides being inversely associated with serum TNF-alpha and IL-12, decreased IL-10 levels were significantly related to increased AHI, hyperinsulinemia, and insulin resistance. Serum IL-10 is significantly reduced in morbidly obese subjects with severe OSA while also showing a clear relationship with a state of hyperinsulinemia and insulin resistance probably regardless of obesity in the present sample. It may be of potential clinical interest to identify the stimulatory mechanisms of IL-10 in obese individuals with OSA.

Summary: This study demonstrates that native LDL (nLDL) synergizes with LPS to alter the balance of human monocyte subsets and their ability to produce inflammatory cytokines and chemokine receptors. Patients with serum LDL-cholesterol (LDL-C) >100 mg/dL showed increased levels of NCM, IL-1 beta, LPS-binding protein (LBP), and Castelli's atherogenic risk index compared to controls with optimal LDL-C concentrations (<= 100 mg/dL).

The article highlights the achievements of the consortium and discusses strategies for prevention and treatment of colorectal cancer in Mexico.

STAT6 controls the stability and suppressive function of regulatory T cells

Summary: Signal transducer and activator of transcription 6 (STAT6) promotes tumorigenesis in colitis-associated cancer (CAC) by decreasing the frequency of Foxp3+ cells and allowing for the infiltration of inflammatory cells. The study found that STAT6 plays a crucial role in the generation of inducible in vitro regulatory T cells (iTregs) and peripheral in vivo Tregs (pTregs) under inflammatory conditions. STAT6 deficiency led to stable iTregs phenotype, increased suppressive ability, and overexpression of PD-1, CTLA-4, and Foxp3, as well as decreased IFN-gamma expression. In a mouse model of CAC, STAT6-/- pTregs exhibited an activated phenotype, higher suppressive capacity, and increased expression of PD-1 and LAP, contributing to the attenuation of tumor development.

Summary: Robust animal data indicate that sucralose intake during pregnancy can lead to weight gain, metabolic disturbances, and low-grade systemic inflammation in offspring, but information in humans is still limited. A cross-sectional, prospective study was conducted to examine the effects of light sucralose ingestion (LSI) or heavy sucralose intake (HSI) during pregnancy on neonatal anthropometric, metabolic, and inflammatory features. Infants from HSI mothers had higher birth weight, insulin levels, and percentage of inflammatory nonclassical monocytes compared to those from LSI mothers. Furthermore, umbilical cord tissue of infants from HSI mothers showed higher levels of IL-1 beta and TNF-alpha with lower IL-10 expression. These results suggest that heavy sucralose ingestion during pregnancy can impact neonatal health.

Summary: This study investigated the incidence of SARS-CoV-2 infection among healthcare workers (HCW) in a dedicated COVID-19 hospital during a six-month follow-up period. The results showed a high infection rate among the HCW cohort, with a majority being asymptomatic carriers. No baseline risk factors contributed to the infection rate. Therefore, it is necessary to enhance hospital safety measures to prevent virus transmission from HCW to relatives and non-COVID-19 patients during future waves of contagion.
