Summary: In this study, the heterogeneity of immune cell subsets during sepsis was analyzed using single-cell RNA sequencing. A unique subcluster of dendritic cells called "mregDC" was identified and its presence and function in the disease were confirmed. Furthermore, the study found that the mregDC program was initiated through specific intracellular signaling pathways within 24 hours of septic challenge.

Summary: This study investigated the serial changes in endotoxin tolerance in a polymicrobial sepsis model. The results showed that endotoxin tolerance started at 6 hours after sepsis, reached its lowest point at 24 to 48 hours, and then started to recover. Endotoxin tolerance was more prominent in severe sepsis.

Summary: The study found that LPS can directly stimulate pDCs via the TLR4/MD2 pathway and promote the activation of pDCs and cDCs. cDCs may contribute to the enhancement of IFN-alpha production in pDCs.

Summary: Astaxanthin has anti-inflammatory effects on murine DCs by regulating cytokine and phenotype marker expression to inhibit inflammation. Additionally, it promotes endocytosis, inhibits DC migration, and reduces T cell proliferation. Finally, it inhibits immune dysfunction in DCs induced by LPS by activating the HO-1/Nrf2 axis, ultimately enhancing survival in LPS-challenged mice.

Summary: Transcripts and proteins of TLR1/2/4/5/6, NOD1/2 and NLRP1/2 were expressed in human periodontal tissue in health and disease, with NOD2 possibly considered as a key damage-associated molecular pattern sensor on periodontal resident cells.

Summary: The absence of mgmt in macrophages resulted in less severe sepsis in the cecal ligation and puncture (CLP) model. However, this protective effect was lost in mice without antibiotics, indicating the importance of microbial control during sepsis immune modulation.

Summary: Severely ill children in low- and middle-income countries experience high mortality rates from infectious diseases, and the risk of infection-related death is compounded by undernutrition. This study found that T cell function is impaired and innate immune responses are exaggerated in acutely ill children. Nutritional status is associated with the magnitude of cytokine responses. Further research is needed to understand the mechanisms of innate immune dysregulation in this population.

Summary: This study investigated the pathophysiological role of BRISC in acute liver failure. The expression of BRISC components was found to increase in kupffer cells (KCs) upon LPS treatment, and the deficiency of BRISC significantly attenuated liver damage and inflammation response. The study demonstrated that BRISC is involved in NF-kappa B-mediated proinflammatory cytokines production in LPS-treated KCs. This research opens the possibility for the development of new strategies to inhibit KCs-driven inflammation in liver diseases.

Summary: In this study, a molecular and supramolecular strategy was reported for developing mixed-charged nanoparticles with resistance to protein adsorption, especially focusing on the creation of invisible capsid-like NPs. The co-assembly of equivalent amounts of anionic and cationic dendritic lipopeptides resulted in neutral mixed-charged NPs, which showed excellent resistance to protein adsorption and served as stealth nanocarriers for harboring guest molecules.

Summary: Chronic lymphocytic thyroiditis, or HD, is a common thyroid disorder. Research has focused on the role of Toll-like receptors (TLRs), particularly TLR2, in the pathogenesis of HD. This study found a significant increase in TLR2 expression on immune cells and a higher concentration of soluble TLR2 in plasma in HD patients compared to healthy individuals. TLR2 expression also correlated with thyroid function indicators. These findings suggest that TLR2 may be involved in the immunopathogenesis of HD.
