Summary: Meiosis is essential for sexual reproduction, with oocytes in female mammals beginning meiosis before birth and sustaining it through dictyate stage before ovulation. Fine-tuning of meiotic progression is crucial for fertility, as any disruption can lead to oogenesis failure and impact overall reproductive health. Understanding the regulatory network of oocyte meiosis is key to comprehending mammalian reproduction and the consequences of misregulation on infertility.

Summary: This study investigates the use of a novel zinc chelator for activating bovine and porcine oocytes. The results show that the zinc chelator can successfully induce blastocyst development in both species, although developmental rates and embryo quality are compromised in bovine compared to calcium-induced embryos. On the contrary, the new concentration and incubation time of the zinc chelator improve developmental rates in porcine without affecting embryo quality. These findings contribute to understanding the role of zinc in oocyte activation and preimplantation embryo development across different mammalian species.

Summary: PABPC1L gene variants are responsible for female infertility characterized by oocyte maturation arrest. These variants result in truncated proteins, reduced abundance, altered localization, and decreased mRNA translational activation. PABPC1L plays an important role in human oocyte maturation and could be a potential candidate gene for infertility causes.

Summary: MICU1 modulators enhance mitochondrial activity, pyruvate metabolism, and developmental competence of thawed oocytes after vitrification. Vitrification leads to adverse effects on mitochondrial function, but these effects can be mitigated by MICU1 activation.

Summary: The force of gravity on Earth has a significant impact on biological processes and tissue functionality. Microgravity, such as in space, has been reported to have negative effects on living beings, including bone demineralization, muscle atrophy, cardiovascular deconditioning, sensory imbalance, altered metabolic and nutritional status, and immune system dysregulation. The use of microgravity simulators, like the Random Positioning Machine (RPM), helps study the effects of gravity variations on human oocytes at the cellular level and reveals that microgravity could compromise oocyte quality by affecting the cytoskeleton, mitochondria localization, and morphology of smooth endoplasmic reticulum (SER). This study suggests that microgravity negatively affects oocyte quality and its competence for fertilization.

Summary: Oocytes are maintained in a state of meiotic arrest until ovulation is triggered. Manipulation of cyclic nucleotides such as cGMP and cAMP can influence meiotic arrest and improve oocyte developmental competence. Pre-IVM modulation of cGMP and/or cAMP has shown consistent improvement in oocyte developmental competence.

Summary: The recent COVID-19 pandemic has brought about significant changes in various fields, including society, law, economics, science, and medicine. The use of mRNA-based vaccines has been approved for the first time by drug regulatory authorities to combat the outbreak. While this represents a novel application of mRNA technology in vaccination medicine, the introduction of RNA into cells to produce molecules is not a new concept. It has been commonly practiced in research to manipulate oocytes and embryos and has potential therapeutic and diagnostic applications in combating infertility. This article describes the potential clinical applications, advantages, and limitations of mRNA-based platforms, and discusses how recent advances driven by the pandemic can benefit infertility treatment. Future directions for enhancing RNA therapeutics in reproductive biology, specifically in oocyte and embryo delivery, are also presented.

Summary: This study compared the developmental competence of oocytes that matured in vivo to metaphase II (MII) with those that matured in vitro from metaphase I (MI). The results showed that MII oocytes had higher fertilization rates, blastocyst formation rates, and euploidy rates compared with MI-MII oocytes. However, there was no significant difference in clinical pregnancy rates, spontaneous pregnancy loss rates, and live birth rates between the two groups after single euploid blastocyst transfers.

Summary: Zinc, an essential trace mineral, plays a critical role in various cellular processes. Zinc deficiency in patients with polycystic ovary syndrome (PCOS) has been found to affect the reproductive system, particularly oocyte quality. This study established an in vitro model to investigate the regulatory function of zinc in porcine oocyte maturation. The results showed that zinc deficiency disrupted meiotic progress and cytoskeleton structure, impaired mitochondrial function, perturbed cytosolic Ca2+ homeostasis, and induced mitophagy in porcine oocytes.

Summary: A homozygous missense mutation (c.895T>C; p.C299R) in TBPL2 was identified in two infertile sisters from a consanguineous family, impairing the transcription initiation function of the protein. The rare and pathogenic TBPL2 mutation may be associated with female infertility due to oocyte maturation arrest and degeneration.
