Dectin-1/Syk signaling is involved in Lactobacillus casei cell wall extract-induced mouse model of Kawasaki disease

Kawasaki disease (KD) is not only the leading cause of childhood acquired heart diseases, but also causes profound coronary artery sequelae due to chronic vascular inflammation in adulthood. Of unknown underlying mechanism, both innate and adaptive immune responses are involved in the pathogenesis of coronary artery lesions (CALs). We investigated the role of dectin-1/spleen tyrosine kinase (Syk) pathway on macrophage in responsive to Lactobacillus casei cell wall extract (LCWE) in vitro and in vivo. We found that LCWE induced in vitro macrophage activation with increased production of IL-6, TNF-alpha, and MCP-1, concomitantly with Syk activation, and dectin-1 and TLR2 enhancement. In vivo, LCWE induced infiltration of dectin-1(+) macrophages into CALs and cardiac upregulation of IL-6 and MCP-1 on day 14 post-injection. Most importantly, Syk inhibition alleviated LCWE-induced arteritis in BALB/c mice. Blockade of either dectin-1 or Syk significantly inhibited LCWE-induced IL-6 and MCP-1 production both in vitro and in vivo. This study demonstrates that the macrophage dectin-1/Syk-mediated pathway is involved in LCWE-induced CALs and production of IL-6 and MCP-1. Given the functional equivalence of human dectin-1 to murine, the importance of dectin-1/Syk pathway in the development of murine CALs warrants further investigation on their roles in human KD (C) 2012 Elsevier GmbH. Summary: This study monitored the dynamic changes in different stages of Kawasaki disease (KD) using near infrared spectroscopy and found that the concentrations of tissue hemoglobin and water showed significant differences. Water content was significantly associated with the formation of coronary artery lesions (CAL). This non-invasive device can help physicians identify KD from fever illness early.

Increased Total Serum Bilirubin Level Post-Ibuprofen Use Is Inversely Correlated with Neonates' Body Weight

Summary: This retrospective study analyzed the data of neonates born at Chang Gung Memorial Hospital in Taiwan and found that the use of ibuprofen resulted in an apparent increase in total serum bilirubin (TSB) level in neonates with lower birth weight. For extremely low birth weight neonates experiencing severe unconjugated hyperbilirubinemia, intravenous acetaminophen can be a suitable alternative to ibuprofen for the treatment of patent ductus arteriosus (PDA).

Gut mycobiome dysbiosis and its impact on intestinal permeability in attention-deficit/hyperactivity disorder

Summary: This study used high-throughput sequencing to explore the dysbiosis of the gut mycobiome in children with ADHD. The results showed that ADHD patients had abnormal fungal community composition and increased abundance of Candida, especially Candida albicans. The in vitro cell assay also demonstrated that C. albicans secretions enhanced intestinal permeability.

General Anesthesia in Early Childhood Significantly Reduces Asthma Incidence and Clinical Visits: A Nationwide Population-Based Cohort Study

Summary: This study examines the correlation between exposure to general anesthesia under three years old and the subsequent course of asthma in children. The study found that the incidence of asthma onset was significantly reduced in children with exposure to general anesthesia under three years old. Furthermore, regardless of the timing of asthma onset, patients with general anesthesia exposure had fewer clinical visits than those without exposure.

Gut Leakage Markers and Cognitive Functions in Patients with Attention-Deficit/Hyperactivity Disorder

Summary: This study investigated the role of gut leakage biomarkers in the susceptibility of ADHD. The results showed that DEFA1 levels were significantly lower in ADHD group compared to the healthy control group. There was no significant difference in serum levels of zonulin and occludin. DEFA1 levels were negatively correlated with inattention scores, while occludin levels were positively correlated with detectability subtest scores. This study provides new insights into the relationship between gut leakage markers and cognition, advancing research on the pathophysiology of ADHD.
