A novel insulin sensitizer (S15511) enhances insulin-stimulated glucose uptake in rat skeletal muscles

Type 2 diabetes is preceded by the presence of skeletal muscle insulin resistance, and drugs that increase insulin sensitivity in skeletal muscle prevent the disease. S15511 is an original compound with demonstrated effects on insulin sensitivity in animal models of insulin resistance. However, the mechanisms behind the insulin-sensitizing effect of S15511 are unknown. The aim of our study was to explore whether S15511 improves insulin sensitivity in skeletal muscles. Insulin sensitivity was assessed in skeletal muscles from S15511-treated rats by measuring intracellular insulin-signaling activity and insulin-stimulated glucose transport in isolated muscles. In addition, GLUT4 expression and glycogen levels were assessed after treatment. S15511 treatment was associated with an increase in insulin-stimulated glucose transport in type IIb fibers, while type I fibers were unaffected. The enhanced glucose transport was mirrored by a fiber type-specific increase in GLUT4 expression, while no improvement in insulin-signaling activity was observed. S15511 is a novel insulin sensitizer that is capable of improving glucose homeostasis in nondiabetic rats. The compound enhances skeletal muscle insulin sensitivity and specifically targets type IIb muscle fibers by increasing GLUT4 expression. Together these data show S15511 to be a potentially promising new drug in the treatment and prevention of type 2 diabetes.

Human and mouse muscle transcriptomic analyses identify insulin receptor mRNA downregulation in hyperinsulinemia-associated insulin resistance

Summary: Hyperinsulinemia, often considered as a compensatory response to insulin resistance, may actually contribute to insulin resistance. The molecular mechanisms underlying this cyclic process are poorly understood. This study found a reliable negative correlation between fasting insulin and INSR mRNA in skeletal muscle. Through cell culture experiments, it was demonstrated that elevated insulin levels attenuated AKT and ERK signaling in muscle cells. RNA-sequencing analysis revealed changes in gene expression related to insulin receptor signaling, FOXO signaling, and glucose metabolism pathways. Hyperinsulinemia led to reduced Insr gene expression and decreased surface INSR protein. SIN3A was identified as a negative regulator of Insr mRNA. These findings provide insights into the mechanisms of hyperinsulinemia-induced insulin resistance in muscle.

HIF1A Knockout by Biallelic and Selection-Free CRISPR Gene Editing in Human Primary Endothelial Cells with Ribonucleoprotein Complexes

Summary: This study reports a highly efficient and selection-free CRISPR gene editing approach using ribonucleoprotein (RNP) complex in primary endothelial cells (HUVECs). The optimized mRNA delivery protocol achieved nearly 100% transfection efficiency of HUVECs with EGFP mRNA. By utilizing this optimized DNA-free approach, highly efficient (98%) and biallelic HIF1A knockout in HUVECs without selection was achieved using three different gRNAs. Functional assays validated the effects of HIF1A knockout on ECs' angiogenic characteristics and response to hypoxia. This work provides a simple method for highly efficient gene editing of primary endothelial cells (HUVECs) in studies and manipulations of ECs functions.
