TIGIT, the Next Step Towards Successful Combination Immune Checkpoint Therapy in Cancer

Summary: TIGIT is an inhibitory receptor expressed on multiple types of lymphocytes, and the efficacy of TIGIT blockade in cancer immunotherapy is currently under investigation. While single TIGIT blockade has limited anti-tumor efficacy, co-blockade with the PD-1/PD-L1 pathway leads to tumor rejection.

Roadmap to next-generation cancer vaccines

Summary: Cancer vaccines have become powerful and clinically viable therapeutic modalities for reducing tumor burden, eradicating residual cancer cells, and preventing relapse. Recent years have seen rapid advancements in next-generation cancer vaccines, with cutting-edge technologies focused on eliciting robust and durable immune responses specific to cancer, as well as reducing systemic side effects. This perspective highlights innovative strategies such as high-throughput screening for neoantigens and optimal delivery systems, knowledge-based de novo design of effective delivery platforms with tissue-targeting specificity, and synergizing cancer vaccines with clinical immunotherapeutic practices like CAR-T and anti-PD-1 therapies.

Nanotechnology for next-generation cancer immunotherapy: State of the art and future perspectives

Summary: Over the past decade, cancer immunotherapy has revolutionized cancer treatment by activating the immune response. However, it is limited by low response rates and toxicity. Nanotechnology offers a promising platform to enhance immunotherapy by improving drug stability, immune cell activation, and safety. Nanotechnology-enabled immunotherapy can target solid tumors, modulate the tumor microenvironment, or alter tumor cell responses to generate effective antitumor immunity. In this Oration, advanced strategies to improve cancer immunotherapy efficacy are discussed along with potential challenges and future directions.

Summary: This review discusses the challenges and future directions of cancer immunotherapies, including monoclonal antibody therapies, adaptive immunotherapies, CAR T cells, T cell attracting bispecific antibodies, TCR mimic antibodies, and new targets. The bispecific T cell attracting monoclonal antibody-mediated killing of cancer cells appears to be the most promising approach for novel effective cancer immunotherapies.

Nanovaccines for cancer immunotherapy: Focusing on complex formation between adjuvant and antigen

Summary: Cancer vaccines have been developed as an interesting cancer immunotherapy approach to deliver tumor antigens and adjuvants to antigen-presenting cells (APCs). Nanotechnology-based vaccines, known as nanovaccines, address the limitations of traditional vaccines by co-delivering antigens and adjuvants to lymphoid organs and APCs, leading to potent anti-tumor immune responses. The formation of nanocomplexes between antigens and adjuvants based on electrostatic interactions has shown promising results in enhancing immune responses. This article summarizes the various platforms for developing cancer vaccines, discusses the advantages of using delivery systems, and focuses on the potential of nanocomplexes formation as cancer nanovaccine platforms.

Targeting tumor-associated macrophage: an adjuvant strategy for lung cancer therapy

Summary: The emergence of immunotherapy has revolutionized cancer treatment, but lung cancer remains a leading cause of death due to resistance. Tumor-associated macrophages (TAMs), a group of immune cells in the tumor microenvironment, play complex roles in lung cancer development. Strategies targeting TAMs have been developed, and their efficacy has been investigated in combination with other treatments. This article summarizes TAMs' mechanisms in lung cancer progression, discusses novel therapeutic strategies, and highlights the potential of TAM targeting as an adjuvant therapy, particularly for improving the efficacy of anti-PD-1/PD-L1 treatment.
