Summary: The multifunctional transcription factor E2f4 plays an important role in zebrafish development, particularly in otolith formation and ciliary beating. Its absence leads to abnormal expression of ciliary motility genes, growth retardation, intestinal microvilli damage, reduced cell proliferation, and increased cell apoptosis. These findings challenge the traditional notion of E2f4 as a transcription repressor.

Summary: SNX17, with high expression in the cardiac outflow tract of rat fetuses, plays a crucial role in cardiac development. Homozygous deletion of SNX17 in rats results in mid-gestational embryonic lethality and congenital heart defects, while heterozygotes exhibit normal fetal development. Additionally, SNX17 deletion leads to abnormal expression of genes associated with cardiac development.

Summary: Sorting nexins (SNXs) are a family of proteins that regulate cellular cargo sorting and trafficking, maintain intracellular protein homeostasis, and participate in intracellular signaling. They also play an important role in the regulation of blood pressure and their aberrant expression and dysfunction can lead to hypertension. Genetic studies have shown a correlation between SNX gene variants and the response to antihypertensive drugs. This review summarizes the progress in SNX-mediated regulation of blood pressure, discusses the potential role of SNXs in the pathophysiology and treatment of hypertension, and proposes novel strategies for the medical therapy of hypertension.

Summary: AQP8 is highly expressed in the gastrointestinal tract of mammals, potentially playing a role in its physiological and pathological processes. In zebrafish, three types of aqp8s are encoded in the genome, with aqp8ab mainly expressed in the embryonic intestine. Knocking out aqp8ab leads to defects in intestine development and abnormal formation of the intestinal lumen.

Summary: This study used zebrafish to investigate the role of TRIC-B in skeletal tissue. The researchers found that the tmem38b gene is expressed at early developmental stages in zebrafish, while tmem38a gene is not. Mutations in tmem38b resulted in under-modified collagen type I and intracellular retention. These findings contribute to our understanding of the importance of TRIC-B in bone cell differentiation.

Summary: Hedgehog (Hh) signaling pathway is highly regulated and mutations in its components can lead to a variety of congenital malformations. Mosmo, a modulator of the Hh pathway, plays a role in down-modulating pathway activation. Studies in zebrafish embryos suggest that MOSMO may be a candidate gene for uncharacterized forms of human congenital craniofacial malformations.

Summary: SNX27 plays a crucial role in cellular sorting processes by associating with retromer for endosomal recycling of transmembrane receptors. Dysregulation of SNX27 is linked to various diseases including Down's Syndrome, Alzheimer's Disease, addiction, and cancer. The potential of SNX27/retromer as a therapeutic target for neurodegenerative diseases is an emerging area of study.

Summary: SNX32 interacts with SNX4 via its BAR domain and with TfR, CIMPR, and BSG through its PX domain, regulating cargo sorting and trafficking processes. Silencing of SNX32 leads to defects in intracellular trafficking and neuronal differentiation, suggesting its crucial role in maintaining neuroglial coordination.

Summary: This study found that FXR regulates BPPC-to-hepatocyte and BPPC-to-BEC redifferentiations through ERK1 and Notch, respectively. FXR agonists are being used in clinical trials for liver diseases, and this study proposes potential underlying mechanisms by characterizing the roles of FXR in the stimulation of dedifferentiation-redifferentiation transition and BPPC redifferentiation.

Summary: The study revealed the crucial role of fgfbp3 in axonal morphogenesis and regeneration of motor neurons in zebrafish, with loss of fgfbp3 leading to defects that can be partially rescued by fgfbp3 mRNA injection and overexpression. Transcriptome sequencing identified drd1b and neurod6a as potential rescuers for motor neuron malformation in fgfbp3 knockout embryos.
