Synergistic Antitumor Effects of Transarterial Viroembolization for Multifocal Hepatocellular Carcinoma in Rats

Oncolytic virotherapy is a promising strategy for safe and effective treatment of malignancy. We have reported previously that recombinant vesicular stomatitis virus (VSV) vectors are effective oncolytic agents that can be safely administered via the hepatic artery in immunocompetent rats to treat multifocal hepatocellular carcinoma (HCC), resulting in tumor necrosis and prolonged survival. Though the results were encouraging, complete tumor regression was not observed, which led us to explore alternative approaches to further enhance the efficacy of VSV treatment. Transarterial embolization techniques have been shown to improve the efficiency and tumor selectivity of anticancer treatments. Degradable starch microspheres (DSM) are one such embolic agent that provides transient embolization of the therapeautic agent before being degraded by serum amylases. Here we demonstrate via dynamic contrast-enhanced magnetic resonance imaging that in our rat model of multifocal HCC, DSM injection into the hepatic artery results in a substantial reduction in tumor perfusion of systemically applied contrast agent. VSV, when administered in combination with DSM, results in enhanced tumor necrosis and synergistically prolongs survival when compared with VSV or DSM monotherapy. Conclusion: This regimen of viroembolization represents an innovative therapeutic modality that can augment the future development of transarterial oncolytic virus therapy for patients with advanced HCC. Summary: This study investigated the potential of high-resolution, high b-value computed DWI in the detection of PDAC. The results showed that high-resolution, high b-value computed DWI significantly improved lesion detection and had a higher incidence of strong lesion-to-pancreas intensity relation compared to conventional high b-value computed and high-resolution high b-value acquired DWI. However, the image quality was rated lower in the high-resolution, high b-value computed DWI datasets.

Summary: Invasive nonfunctioning pituitary neuroendocrine tumors (PitNETs) are difficult to remove and current therapies often fail. This study investigated the ANGPT2/TIE2 axis in NF-PitNETs and found that PitNET cells express functional TIE2 receptors and secrete bioactive ANGPT2, which promotes tumor cell growth. Inhibition of this signaling pathway showed promise in inhibiting PitNET growth in various models. Therefore, the ANGPT2/TIE2 axis could be a potential therapeutic target for NF-PitNETs.

Noise reduction in diffusion weighted MRI of the pancreas using an L1-regularized iterative SENSE reconstruction

Summary: The study evaluated the use of L1 regularized iterative SENSE reconstruction (L1-R SENSE) to improve image quality and robustness of ADC estimation in high resolution diffusion-weighted magnetic resonance imaging of the pancreas. Compared to traditional SENSE reconstruction, L1-R SENSE resulted in higher image quality, reduced noise artifacts, and improved ADC measurements for both PDAC and normal pancreas. Although one reader found SENSE to be superior in anatomical details, overall L1-R SENSE performed better in terms of noise reduction and robustness in ADC estimation.

Summary: The study aimed to assess the impact of respiratory motion and cardiac driven pulsation on renal DWI and investigate the use of asymmetrical velocity-compensated diffusion encoding waveforms for robust ADC mapping in the kidneys. The results showed that short trigger delays in respiratory-triggered scans could cause higher ADC values, especially in the right kidney. The asym-vc waveform was found to reduce ADC variation due to respiratory motion, although at the cost of reduced SNR compared to the pgse waveform.

Algorithmic transparency and interpretability measures improve radiologists' performance in BI-RADS 4 classification

Summary: This study evaluated the perception of different types of AI-based assistance and the interaction of radiologists with the algorithm's predictions and certainty measures. Four radiologists classified Breast Imaging-Reporting and Data System 4 (BI-RADS4) lesions and the effect of different AI-based assistance on their performance was measured. The results showed that trust in the algorithm's performance was mostly dependent on the certainty of the predictions in combination with a plausible heatmap, and the human-AI interaction varied widely and was influenced by personality traits.

Summary: There is a strong correlation between trabecular vBMD values in the cervicothoracic and lumbar regions, with vBMD values decreasing with age in both women and men. The proposed cut-off values for osteoporosis peaked at C4 and decreased towards T12, allowing for the determination of low bone mass in clinical cases where only parts of the spine are imaged.
