Summary: Vascular ageing is closely related to atherosclerosis and enhanced vascular inflammation. Phospholipid oxidation products induce cellular inflammation and affect cell behavior through interactions with vascular cells and immune cells. Interventions in lipid metabolism and anti-inflammation are important pathways for treating atherosclerosis. This review aims to sort out the signaling pathway of oxidized phospholipids-induced inflammatory factors and changes in cell behavior, and summarize therapeutic targets for the development of atherosclerosis drugs.

Summary: The accumulation of 25-hydroxycholesterol (25-HC) in human coronary atherosclerotic lesions accelerates atherosclerosis progression and promotes plaque instability through autocrine and paracrine actions. Macrophage-derived 25-HC intensifies the inflammatory response of lipid-loaded macrophages and inhibits smooth muscle cell migration within the plaque. These effects are independent of 25-HC-mediated modulation of transcriptional activity.

Summary: Atherosclerosis is a chronic disease with inflammatory components, now recognized as an active autoimmune process involving both innate and adaptive immune pathways. One of the key factors driving this process is the presence of modified LDL, which leads to the formation of oxidation-specific epitopes recognized by immune cells. Macrophage response to these epitopes is crucial for initiating and promoting inflammation in the arteries.

S100a9 is identified as a pivotal mediator in the protective effects of melatonin, and its overexpression can abolish the antagonistic effect of melatonin on HFD-induced vascular inflammation.

Summary: Gpx4 and Alox15 are enzymes involved in oxidative lipid metabolism, and play important roles in spermatogenesis. This study found that Gpx4 is widely distributed in mouse sperm and its deficiency affects acrosomal exocytosis and in vitro fertilization. Alox15 deficiency had minimal impact on acrosome reactions, but induced spontaneous acrosomal exocytosis when crossed into a Gpx4-deficient background.
