Assessing the clinical value of faecal bile acid profiling to predict recurrence in primary Clostridioides difficile infection

Benjamin H. Mullish, Laura Martinez-Gili, Elena Chekmeneva, Goncalo D. S. Correia, Matthew R. Lewis, Verena Horneffer-Van Der Sluis, Lauren A. Roberts, Julie A. K. McDonald, Alexandros Pechlivanis, Julian R. F. Walters, Emma L. McClure, Julian R. Marchesi, Jessica R. Allegretti

Summary: This study aimed to explore the dynamics of fecal bile acids (BAs) in patients with primary Clostridioides difficile infection (CDI) and investigate their potential for predicting recurrence. The results showed that gut BA metabolism dynamics differed between patients with and without recurrence, suggesting that individual BAs may serve as novel biomarkers for predicting CDI recurrence.

Patients with Inflammatory Bowel Disease Have Heterogeneous Treatment Preferences That Are Largely Determined by the Avoidance of Abdominal Pain and Side Effects [P-POWER IBD Study]

Edouard Louis, Corey A. Siegel, Barbara James, Sebastian Heidenreich, Nicolas Krucien, Subrata Ghosh

Summary: This study aimed to identify the benefit-risk trade-offs that IBD patients are willing to make in order to inform patient-centric treatment targets. The study found that efficacy, administration, and safety all had varying degrees of impact on treatment preferences. Factors such as abdominal pain, risks of mild-to-moderate side effects, and serious infections were considered important. Preferences varied among patients, with CD patients placing greater importance on abdominal pain and UC patients on bowel urgency.

Wolf, Michael Chiorean, Severine Vermeire, Anjali Jain, Lorna Charles, Garrett Lawlor, Mark T. Osterman, Hsiuanlin Wu, James B. Canavan, Annkatrin Petersen, Jean-Frederic Colombel, Miguel Regueiro

Summary: This interim analysis examines the efficacy and safety of ozanimod treatment in patients with ulcerative colitis over a period of approximately 3 years. The results show that a high percentage of patients maintain clinical and mucosal efficacy during the treatment. No new safety concerns arise from long-term use of ozanimod.

Summary: This study evaluated the correlation and predictive value of a new and established index in ulcerative colitis. The results showed that PHRI had a similar correlation with endoscopy and was able to effectively stratify the risk of relapse. This study demonstrates that a neutrophil-only assessment is a simple and viable alternative.

Practical Management of Biosimilar Use in Inflammatory Bowel Disease (IBD): A Global Survey and an International Delphi Consensus

Ferdinando D'Amico, Virginia Solitano, Fernando Magro, Pablo A. Olivera, Jonas Halfvarson, David Rubin, Axel Dignass, Sameer Al Awadhi, Taku Kobayashi, Natalia Sousa Freitas Queiroz, Marta Calvo, Paulo Gustavo Kotze, Subrata Ghosh, Laurent Peyrin-Biroulet, Silvio Danese

Summary: As the patents for biologic originator drugs expire, biosimilars are emerging as cost-effective alternatives within healthcare systems. A global survey was conducted to shed light on physicians' knowledge, beliefs, practical approaches, and concerns related to biosimilar adoption. Based on the survey results, a consensus was reached on key statements covering biosimilar effectiveness, safety, indications, rationale, multiple switches, therapeutic drug monitoring, non-medical switching, and future perspectives. The consensus affirmed that biosimilars are equally effective and safe compared to originator drugs.
