Summary: The proneural transcription factor Ascl1 is involved in neural fate decisions. It has been widely used to reprogram non-neuronal cells into neurons in vitro. However, this study found that, in vivo, Ascl1 only had very low efficiency in inducing neuronal reprogramming of glial cells in the postnatal mouse cerebral cortex. Instead, Ascl1 selectively enhanced the proliferation of oligodendrocyte progenitor cells (OPCs).

Summary: This study investigated the activation and expression of CRTC isoforms in rat adrenal glands under different stress conditions. The results showed that CRTC2 and CRTC3 play important roles in the transcriptional regulation of StAR, with their activation and nuclear localization associated with changes in plasma ACTH levels. Both isoforms were found to bind to the Star promoter, with CRTC3 binding dynamics corresponding to changes in nuclear protein levels. While little variation was observed in CRTC isoform expression over a 24-hour period, long-term down-regulation was observed following endotoxic stress.

Summary: Sorafenib is a multikinase inhibitor with antitumour effects related to various biological processes, such as proliferation, migration and invasion. Research has shown that sorafenib can target the MEK5/ERK5 signalling pathway, inhibiting ERK5 phosphorylation and affecting transcriptional activation, leading to effects on cell cycle progression and migration in response to EGF. This establishes a new mechanistic insight for the antitumour effect of sorafenib.
