Timing is everything: stochastic origins of cell-to-cell variability in cancer cell death

The diversity of cell populations is regulated by extracellular and intracellular variability. The latter includes genetic, epigenetic and stochastic variability, all contributing to the experimentally observed heterogeneity in response to external death-inducing stimuli. Studies of sources and regulation of variability in commitment to apoptotic cancer cell death are likely to identify the fundamental features of apoptotic protein networks that are responsible for determining the ultimate cell fate. Systems biology approaches, involving computer simulations of the biochemical reactions accompanied, if possible, by experimental verification of selected components of the model, are proving useful in determining the origins of cell-to-cell variability in response to external stress stimuli. Here we summarize our current understanding of the origins of stochastic variability in cells' commitment to apoptosis, and its implications in the field on cancer therapy.

Summary: Neurotoxicity effects of industrial contaminants are currently not well studied, with behavioral changes in small aquatic invertebrates being proposed as useful indicators, but limited by lack of advancements in analytical technologies. Advances in Lab-on-a-Chip technologies show promise in addressing these limitations and facilitating high-throughput neuro-behavioral ecotoxicity testing.

Summary: Cell adhesion molecules play a crucial role in nervous system development and function. This study identifies NRCAM variants associated with a neurodevelopmental disorder and provides evidence of the detrimental effects on NRCAM structure and function. High-Throughput Phototactic Ecotoxicity Biotests with Nauplii of Artemia franciscana

Summary: Analysis of sensorimotor behavioral responses to stimuli such as light can provide enhanced relevance in prioritizing chemical risks. However, there have been minimal studies on using invertebrate phototactic behaviors in aquatic ecotoxicity testing due to technical limitations. In this research, a high-throughput phototactic biotest system for brine shrimp nauplii was demonstrated, along with a novel bioinformatic approach for analyzing phototactic behaviors at scale. The preliminary data showed consistent light-seeking behaviors in nauplii without habituation, and exposure to sub-lethal concentrations of insecticides resulted in perturbed light seeking behaviors. These findings suggest that phototactic bioassays have the potential to be a sensitive, responsive, and cost-effective method in behavioral ecotoxicology.

Sex differences in the predictability of risk-taking behavior

Summary: Recent research suggests that individuals vary in the consistency of their behavior, and that these differences may be heritable. This study investigated whether there are sex differences in the predictability of risk-taking behavior in male and female eastern mosquitofish. The results showed consistent sex differences in mean-level risk-taking behavior, but the magnitude and direction of sex differences in behavioral predictability were assay-specific.

Toward Real-Time Animal Tracking with Integrated Stimulus Control for Automated Conditioning in Aquatic Eco-Neurotoxicology

Summary: Aquatic eco-neurotoxicology is a new field that focuses on studying the effects of pollutants on animal behaviors, particularly the cognitive behaviors affected by neurotoxicants. However, the scarcity of experimental data is due to the lack of low-cost technologies for analyzing higher-level neurological functions in small aquatic organisms. In this study, a proof-of-concept prototype was presented, utilizing real-time animal tracking software and closed-loop external hardware for delivering stimuli based on specific behaviors. The system demonstrated precise animal tracking across different aquatic species and has potential applications in cognitive research of small aquatic model organisms.
