To investigate whether diabetes-induced alterations of CYP2E1 and oxidative stress can be modulated by dietary taurine supplementation, male Wistar rats were divided into non-diabetic, diabetic, and diabetic taurine-supplemented groups (administered at 2% in the drinking water). Increased levels of CYP2E1-catalyzed p-nitrophenol hydroxylation were found in liver and kidney microsomes of rats with ST-Z-induced diabetes compared to those of non-diabetic control rats. Immunoblot and RT-PCR analyses of CYP2E1 protein and mRNA levels in the liver and kidneys showed the same trend as with enzyme activities. Taurine supplementation significantly decreased the enzyme activity and expression (protein and mRNA) of CYP2E1 in diabetic rat kidneys. Plasma beta-hydroxybutyrate concentration was significantly reduced in taurine-treated diabetic rats. The induction of heme oxygenase-1 mRNA was suppressed by taurine treatment in diabetic rat kidneys. An increase in reduced glutathione (GSH) and a higher ratio of reduced to oxidized glutathione (GSH/GSSG) together with lower values of thiobarbituric acid-reactive substances (TBARS) were observed in the kidneys of taurine-treated diabetic rats. However, taurine supplementation caused only a slight or insignificant effect on these alternations in the liver of diabetic rats. Our results show dietary taurine may reduce CYP2E1 expression and activity, and oxidative stress in kidneys of diabetic rats. (C) 2009 Elsevier Ltd. All rights reserved.

Ambient Particulate Matter Induces Vascular Smooth Muscle Cell Phenotypic Changes via NOX1/ROS/NF-κB Dependent and Independent Pathways: Protective Effects of Polyphenols

Chia-Chi Ho, Yu-Cheng Chen, Ming-Hsien Tsai, Hui-Ti Tsai, Chen-Yi Weng, Shaw-Fang Yet, Pinpin Lin

Summary: Epidemiological studies have shown a link between ambient particulate matter exposure and vascular diseases. The research revealed that exposure to ambient PM led to increased cell migration ability in vascular smooth muscle cells and pulmonary arterial smooth muscle cells. Additionally, the study demonstrated that PM induced intracellular reactive oxygen species, activated the NF-kappa B signaling pathway, and increased expression of proinflammatory cytokines in VSMCs. Polyphenolic compounds like curcumin, resveratrol, and gallic acid were found to prevent PM-induced migration by blocking the ROS-dependent NF-kappa B signaling pathway in VSMCs.

An integrated strategy by using long-term monitoring data to identify volatile organic compounds of high concern near petrochemical industrial parks

Chin-Yu Hsu, Pei-Yu Wu, Yu-Cheng Chen, Pau-Chung Chen, Yue Leon Guo, Yi-Jun Lin, Pinpin Lin

Summary: This study analyzed comprehensive measurement data of 109 VOCs collected from 17 monitoring stations around petrochemical industrial parks in Taiwan from 2015 to 2018. The results identified six VOCs, including 1,3-butadiene, benzene, 1,2-dichloroethane, chloroform, vinyl chloride, and acrylonitrile, with more than 50% of their emissions attributed to petrochemical industrial parks. This integrated approach can assist regulatory agencies in proposing effective control strategies to reduce the health risk associated with high-concern VOC emissions.

Worldwide serum concentration-based probabilistic mixture risk assessment of perfluoroalkyl substances among pregnant women, infants, and children

Kuan-Yu Kuo, Yu Chen, Yi Chuang, Pinpin Lin, Yi-Jun Lin

Summary: This study investigated the health risks of perfluoroalkyl substances (PFASs) on pregnant women, infants, and children, with a focus on the mixture risks of the four main PFASs. The findings revealed that PFOS was the most dominant PFAS in the serum of pregnant women and children globally, with potential immune effects exceeding 1. The cumulative hazard quotients (HQs) for developmental and immune effects in infants could also be greater than 1, suggesting concerns for infant development and immunity. The national and global serum database and risk assessment provide additional insights into PFAS exposure and mixture risks in vulnerable populations, serving as a reference for evaluating regulatory mitigation measures.
