Summary: This study analyzed the association between various alterations in sperm morphology and DNA damage level, PRM deficiency, and the abundance of PRM1 gene and protein in sperm. The results showed that each parameter is correlated with one another, indicating the essential role of gene expression and PRM1 protein abundance in sperm chromatin stability, especially regarding DNA damage and sperm head morphology alterations. Additionally, the study demonstrated the effectiveness of CMA3 as an alternative test for detecting PRM content in sperm, without the need for molecular-level assessments.

Summary: The study aimed to investigate the relationship between DNase activity associated with bacterial contamination of incubated bovine frozen-thawed spermatozoa and elevated sperm DNA fragmentation. The experiments showed that incubation of plasmid with the supernatant of infected bovine semen straws resulted in clear evidence of DNase activity compared to non-infected supernatant. The presence of bacteria in semen straws led to an exponential increase in bacterial growth and sperm DNA fragmentation, highlighting the importance of hygiene practices during semen processing.

Summary: Despite the widespread use of sperm cryopreservation, it can cause detrimental alterations in sperm function, including decreased viability, motility, and DNA fragmentation. Oxidative stress appears to play a prominent role in the mechanisms of human sperm cryodamage. Antioxidants such as myo-inositol have been shown to minimize the detrimental effects of reactive oxygen species (ROS) and improve sperm quality and motility. This study demonstrated that post-thaw addition of myo-inositol significantly improved sperm parameters, increased oxygen consumption, and decreased carbonyl groups, indicating reduced oxidative stress. These findings suggest that the developed sperm cryopreservation procedure may improve current in vitro techniques used for sperm cryostorage in assisted reproductive technology (ART) laboratories.

Summary: The study aimed to evaluate the benefits of MitoQ in rooster semen extenders on sperm quality, motility parameters, antioxidant capacities, and apoptotic changes. The results showed that MitoQ at a concentration of 150 nM improved post-thawed sperm quality and motility by restoring ATP levels and preventing membrane damage. It also improved redox balance and exhibited antiapoptotic activity. These findings suggest that MitoQ can be an effective additive in rooster semen cryopreservation technology.

These findings provide insights into the underlying processes of proteome alterations and sperm freezing resistance, contributing to further research on sperm cryopreservation.
