Summary: Clearance-driven optimization was conducted to improve the NIK inhibitor by utilizing metabolite identification studies and structure-based drug design. This resulted in the discovery of an azabicyclo[3.1.0]hexanone motif that reduced in vitro and in vivo clearance, maintained NIK potency, increased selectivity over other kinases, and achieved a more than ten-fold reduction in predicted human dose.
