Induction of virulence factors, apoptosis, and cytokines in precision-cut hamster liver slices infected with Entamoeba histolytica

Precision-cut liver slices (PCLS) are mainly used to evaluate hepatotoxicity and metabolism of chemicals, as well as to study mechanisms of liver damage and repair. However, recently they have been used as a system to study amoebic infections. The aim of this study was to validate this model as an alternative for experimental amoebic liver absess (ALA) in animals. To do this, the PCLS was analyzed for the expression of amoebapore and cysteine proteinases 1 and 5, three of the most studied virulence factors of Entamoeba histolytica, as well as the induction of apoptosis and cytokines production in response to the ex vivo infection. PCHLS were prepared with the Brendel-Vitron tissue slicer and then, infected with 200,000 trophozoites of E. histolytica. Samples were taken at 0, 6, 12, 18, and 24 h and compared to control non-infected slices. Morphological studies were performed in order to verify the infection; while apoptosis was studied by TUNEL and PAS techniques. The expression of cysteine proteinases (1 and 5), and amoebapore, was analyzed by real-time PCR. By using ELISA assays, the production of cytokines was also studied. PCHLS were found to be a reproducible infection system, and E. histolytica caused the expression of cysteine proteinases and amoebapore in infected slices. At the same time, trophozoites induce release of cytokines and apoptotic death of the hepatocytes close to them. PCHLS represent a new and suitable alternative model to study the pathogenesis of hepatic amoebiasis. (C) 2012 Elsevier Inc. All rights reserved.

Summary: This study identified breast cancer-associated genes in young, Mexican women through genetic analysis. The results revealed both well-known genes and rarely reported variants in this population. Furthermore, candidate causal genes were successfully assigned to a significant portion of patients, which could contribute to improved diagnosis, treatment, and life expectancy in young women with breast cancer.

Genomic Characterization by Whole-Exome Sequencing of Hypermobility Spectrum Disorder

Summary: Currently, there is no established genetic basis to differentiate hypermobility spectrum disorders (HSD) from hypermobile Ehlers-Danlos syndrome (hEDS). This study used whole-exome sequencing to identify DNA mutations in patients diagnosed with generalized HSD. Three genes (MUC3A, RHBG, and ZNF717) were found to be mutated in all five patients evaluated. Functional enrichment analysis revealed the extracellular matrix (ECM) structural constituent as the primary overrepresented molecular function. Comparison with other studies supported the hypothesis of two distinct diseases sharing a common genetic background. Five potential biomarkers for HSD were identified.

Biological Activities of Seven Medicinal Plants Used in Chiapas, Mexico

T. nelsonii exhibited the highest antimicrobial and antihemolytic activities, suggesting its potential as a source of compounds for cancer treatment and blood sugar control.

Laurequinone, a Lead Compound against Leishmania

Summary: In this study, it was found that five compounds isolated from the red alga Laurencia johnstonii exhibited leishmanicidal activity. Among them, laurequinone showed the strongest activity and was capable of inducing programmed cell death or apoptosis in the parasite. These findings highlight the potential of laurequinone as a novel therapeutic agent against kinetoplastid parasites.

Antimycobacterial Activity of Hedeoma drummondii against Mycobacterium tuberculosis and Non-Tuberculous Mycobacteria

Summary: Tuberculosis (TB) is still a major global health problem, with antibiotic-resistant strains posing the greatest challenge to its treatment. Additionally, the prevalence of non-tuberculous mycobacteria (NTM) in humans has significantly increased in recent years. This study aims to investigate the antimycobacterial effects of extracts and major compounds from Hedeoma drummondii on both clinical isolates of Mycobacterium tuberculosis and NTM strains. The methanolic extract exhibited the best activity against M. tuberculosis, inhibiting 10 out of 12 strains at a concentration < 2500 mu g/mL, while the hexanic extract showed the best activity against NTM, inhibiting 8 out of 10 strains at <= 625 mu g/mL. Furthermore, there was a strong positive correlation between the antimycobacterial activity of pulegone and the hexanic extract against NTM strains, suggesting its potential as a predictive marker for these microorganisms.
